High expression of α-SMA and type Ⅰ collagen induced by transcription factor T-bet/GATA-3 ratio in asthma models

Abstract: ObjectiveTo study the relationship between transcription factor T-bet/GATA -3 and expression of α-smooth muscle actin (α-SMA) and type Ⅰ collagen in asthma. MethodsThe levels of T-bet and GATA-3 in rat lungs were determined by quantitative real-time PCR and western blotting respectively. α-SMA and type Ⅰ collagen levels in lung fibroblasts were determined by immunohistochemistry and ELISA respectively. ResultsThe expression of T-bet mRNA and protein in the OVA group was significantly attenuated compared with that in the saline group and the blank group, while the expression of GATA-3 in the OVA group was significantly increased and the T-bet/GATA-3 ratio in the OVA group was significantly decreased. The α-SMA and type Ⅰ collagen levels in the OVA group were significantly increased. The T-bet/GATA-3 ratio had a negative correlation with α-SMA and type Ⅰ collagen. There was no significant difference between the saline group and the blank group. ConclusionThe T-bet/GATA3 ratio has a negative relation to α-SMA and type Ⅰ collagen, and T-bet/GATA3 imbalance plays a crucial role in asthma models.
