Antimicrobial effect of diallyl trisulfide on Helicobacter pylori biofilms

摘要:

目的  探讨二烯丙基三硫醚(DATS)对幽门螺杆菌生物膜的杀菌作用,进一步研究生物膜耐药的机制。 方法微量肉汤法测定药物的最小抑菌浓度,平板培养法构建幽门螺杆菌生物膜的体外模型,XTT法检测药物处理后浮游状态与生物膜状态下的幽门螺杆菌活性,激光共聚焦显微镜观察生物膜深层的细菌状态以及活性变化。 结果克拉霉素与DATS对浮游状态的幽门螺杆菌均有良好的杀菌作用;使用不同浓度克拉霉素处理后的幽门螺杆菌生物膜活性明显低于对照组;DATS处理后的幽门螺杆菌生物膜活性下降明显,与对照组和克拉霉素组有统计学差异。结论  DATS对浮游状态和生物膜状态下的幽门螺杆菌均具有良好的杀菌活性,可以为耐药菌临床治疗提供参考。

Abstract:

ObjectiveTo investigate the antimicrobial effects of diallyl trisulfide (DATS) on Helicobacter pylori(H.pylori) biofilms and to explore the drug-resistance mechanism of biofilms. Methods  The minimal inhibitory concentration of selected antimicrobials were measured by broth microdilution assay; The biofilm models in vitro were established on agar plate; The cell activity of planktonic and sessile H.pylori treated with selected antimicrobials was measured with XTT reduction assay; The morphology and activity changes of H.pylori in deep biofilm were observed by confocal laser scanning microscopy. Results  Both clarithromycin and DATS elicited strong antimicrobial activity against planktonic and sessile H.pylori; Different concentrations of clarithromycin significantly reduced the cell activity of H.pylori biofilm; The cell activity of the DATS treated group was significantly lower than that of the control group and the clarithromycin treated group. It had statistical differences. Conclusion  DATS elicits strong antimicrobial activity against both planktonic and sessile H.pylori. It may act as an alternative chemotherapeutic agent for multidrug-resistant bacterial strains.
