三叉神经痛大鼠脑内P物质及β-内啡肽的变化

Changes of substance P and betaEndorphin in the brain of  rats with trigeminal neuralgia

摘要:

目的   建立大鼠三叉神经痛模型,测定其下丘脑、垂体P物质(SP)及β-内啡肽(β-EP)含量的变化,进一步探讨三叉神经痛的发病机制。方法   将28只SD成年雄性大鼠随机分为实验组(A组)、生理盐水组(B组)、对照组(C组)。A组:脑闩下蛛网膜下腔注入3μL青霉素G-K,三叉神经痛模型建立后,处死大鼠,取下丘脑、垂体,按放射免疫法处理,同批测定SP及 β-EP的含量。B组:脑闩下蛛网膜下腔注入3μL生理盐水,30min后取下丘脑、垂体,余同A组。C组:直接处死,余同A组。结果   实验组SP含量明显高于生理盐水组及对照组(P<0.01),而β-EP含量实验组明显低于生理盐水组和对照组(P<0.01), 生理盐水组与对照组SP和β-EP含量差异无统计学意义(P>0.05)。结论   神经肽SP和β-EP在三叉神经痛的发病中可能起重要作用。

Abstract:

Objective   To explore the pathogenesis of trigeminal neuralgia by establishing a rat model of trigeminal neuralgia and measuring changes of SP and β-EP in hypothalamus and hypophysis. Methods   Twenty-eight male adult Sprague-Dawley rats were randomly assigned to the experimental group(Group A), the Sodium Chloride group(Group B) and the control group(Group C). In Group A, the rats were injected with 3μL penicillin G-K into the subarchnoid space caudal to obex. After the models were established, they were killed, and then their hypothalamus and hypophysis were taken out and were determined for the concentrations of SP and β-EP by radioimmunological assay(RIA) . As to Group B, the progress was the same as Group A except that the rats were injected with 3μL Sodium Chloride and were killed 30 minutes later. And the rats in Group C were directly killed, others just following Group A. Results   The concentration of SP in the experimental group was significantly higher than that in the Sodium Chloride group and the control group (P<0.01),while the concentration of β-EP was much lower(P<0.01) but there was no considerable difference between the Sodium Chloride group and the control group (P>0.05).  Conclusion   Neuropeptides, such as SP and β-EP, may play important roles in the pathphysiological mechanism of trigeminal neuralgia.
