摘要: 目的 探讨子宫内膜癌肿瘤微环境及转化生长因子β-1(TGF β-1)对正常间质细胞(NF)活化为肿瘤相关成纤维细胞(CAF)的影响。 方法 (1)体外培养人子宫内膜癌细胞系Ishikawa细胞,制备条件培养基(CM),原代培养NF,形成共培养体系,Western blotting与QT-PCR方法检测其α-平滑肌肌动蛋白(α-SMA)的表达。(2)分组为:A:NF,B:NF+TGF β-1,C:NF+CM,D:NF+CM+TGF β-RⅠ/Ⅱ激酶抑制剂。 结果 B和C组α-SMA蛋白表达较A组高(P=0.01、P<0.001), B和C组α-SMA的mRNA表达较A组高,两组比较差异有统计学意义(P<0.001、P=0.01);B和C组α-SMA蛋白表达较D组高(P<0.001、P=0.01),B和C组α-SMA的mRNA表达较D组高,两组比较差异有统计学意义(P<0.001、P=0.01)。与C组相比,B组α-SMA蛋白表达较C组高(P<0.001)。B组与C组比较α-SMA mRNA表达差异无统计学意义(P=0.40)。 结论 子宫内膜癌肿瘤微环境可促进NF-CAF的转化,TGFβ-1在该过程中发挥重要作用。
