洋葱总黄酮对大鼠糖尿病视网膜神经节细胞的神经保护作用

Neuroprotective effect of flavanoids extracted from onion on retinal ganglion cells with diabetes mellitus

摘要: 目的 考察糖尿病大鼠视网膜神经节细胞(RGCs)的凋亡及凋亡相关基因的表达,探讨洋葱总黄酮对 RGCs 的神经保护作用。 方法 50只雄性SD大鼠分为对照组,糖尿病模型组(DM组),洋葱总黄酮小剂量组(Ⅰ组)、中剂量组(Ⅱ组)和大剂量组(Ⅲ组),每组10只。Ⅰ组、Ⅱ组和Ⅲ组分别按50、100、200 mg/kg剂量每日给予洋葱总黄酮灌胃1次。TUNEL法检测 RGCs 凋亡并计数凋亡数量。LSAB 法检测 Bcl-2、Bax在 RGCs层的表达。 结果 与对照组相比,DM组大鼠血糖升高,体质量下降(P<0.01),视网膜中的Bcl-2和Bax表达增强,RGCs凋亡细胞数量显著增加(P<0.01);与DM组相比,Ⅱ组和Ⅲ组血糖降低(P<0.01),Bcl-2表达增强,Bax表达减弱(P<0.05,P<0.01),RGCs凋亡细胞数量显著减少(P<0.01),Ⅲ组体质量增加(P<0.05)。 结论 洋葱总黄酮能促进 Bcl-2 的表达,抑制Bax的表达,减少DM组RGCs的凋亡,对RGCs具有神经保护作用,从而对糖尿病视网膜病变起一定的防治作用。

Abstract: Objective To detect the apoptosis of the retinal ganglion cells(RGCs)in rats with diabetes mellitus and expression of the apoptosis-related genes, then to explore the neuroprotective effect of flavanoids extracted from onion on RGCs. Methods Diabetic rats were induced by streptozotocin. Fifty rats were randomly divided into five groups: normal control group, diabetic model group(DM group), low-dose flavanoids extracted from onion group(I group), medium-dose flavanoids extracted from onion group(II group), and high-dose flavanoids extracted from onion group(III group), ten in each group. Rats in I, II and III groups were treated with flavanoids extracted from onion once each day. The apoptosis of RGCs were measured by TUNEL assay. The expression levels of Bcl-2 and Bax were measured by LSAB. Results Compared with normal control group, the blood sugar of rats in DM group were significantly increased, the body weight were significantly reduced(P<0.01), expression levels of Bcl-2 and Bax were enhanced, and the numbers of apoptosis RGCs were increased significantly(P<0.01). Compared with DM group, the blood sugar of rats in II and III groups were significantly decreased(P<0.01), expression level of Bcl-2 was enhanced but that of Bax was decreased(P<0.05, P<0.01), the numbers of apoptosis RGCs were decreased significantly(P<0.01), and the body weight of rats in III group were significantly increased(P<0.05). Conclusion Flavanoids extracted from onion can induce the expression of Bcl-2 while inhibit that of Bax, reduce the apoptosis of RGCs, and protect RGCs neurologically.
