HMGB1与MMP-9在早期胃癌患者血清中的表达及其临床意义

Expression of serum HMGB1 and MMP-9 in early gastric cancer patients and its clinical significance

摘要:

目的  探讨血清高迁移率族蛋白B1(HMGB1)和基质金属蛋白酶-9(MMP-9)在早期胃癌中的表达及临床意义。 方法应用酶联免疫吸附法(ELISA)检测92例早期胃癌患者血清HMGB1和MMP-9水平,并与40例胃良性病变患者和30例健康对照者比较。 结果胃癌组患者血清HMGB1与MMP-9水平明显高于胃良性病变组和健康对照组(P均<0.05),而胃良性病变组与对照组差异不明显(P>0.05);相关分析显示,胃癌患者血清HMGB1和MMP-9水平之间呈正相关(P<0.01),血清HMGB1和MMP-9的表达水平与淋巴结转移和胃癌的分化程度密切相关(P<0.05),胃癌患者手术后HMGB1和MMP-9水平较手术前明显下降(P<0.05)。 结论HMGB1和MMP-9与早期胃癌的发生发展有关,检测血清HMGB1和MMP-9水平对于早期胃癌的辅助诊断、疗效监测及预后判断具有重要的临床意义。

Abstract:

Objective  To evaluate the value of serum high-mobility group box-1(HMGB1) and matrix metalloproteinase-9(MMP-9) expressions in the diagnosis of early gastric cancer(EGC) and its clinical significance. Methods  92 patients with EGC,40 patients with benign gastric diseases and 30 healthy controls were recruited in this study. Then the HMGB1 and MMP-9 levels in the serum of all the subjects were measured by ELISA respectively and compared. Results  The levels of serum HMGB1 and MMP-9 in the EGC group were significantly higher than those in benign gastric diseases and healthy controls (all P<0.05),   while no  obvious difference in the levels of serum HMGB1 and MMP-9 was observed between benign gastric diseases and healthy controls (P>0.05); the correlation analysis showed that the HMGB1 level was positive correlated with the MMP-9 level (P<0.01); the levels of serum HMGB1 and MMP-9 were significantly associated with lymph node metastasis and tumor differentiation (P<0.05); the HMGB1 and MMP-9 levels were significantly lower in EGC after surgery (P<0.05). Conclusion  HMGB1 and MMP-9 play a role in the tumorigenesis and development of EGC.Measuring of serum HMGB1 and MMP-9 may be a valuable tool in diagnosis, effect monitoring and prognosis in EGC.
