HO-1对AngⅡ诱导的乳鼠心肌细胞凋亡的作用

Role of Heme oxygenase-1 on the apoptosis of myocardial cells induced by angiotensin Ⅱ

摘要:

目的  探讨血红素氧合酶-1(HO-1)对血管紧张素Ⅱ(AngⅡ)诱导的乳鼠心肌细胞凋亡的作用。方法   原代培养1~2d Wistar乳鼠心肌细胞,随机分为对照组、AngⅡ组(加入AngⅡ)、HO-1诱导组(加入AngⅡ和HO-1诱导剂氯化血红素hemin)和HO-1抑制组(加入AngⅡ和HO-1抑制剂锌原卟啉-9 ZnppIX)。采用Western blotting检测心肌细胞HO-1蛋白的表达,Hoechst及流式细胞仪Annexin-V/PI检测细胞凋亡。结果   AngⅡ组较对照组心肌细胞凋亡率明显升高(P<0.05);HO-1诱导组较AngⅡ组细胞凋亡率明显下降(P<0.05),HO-1蛋白表达水平明显升高(P<0.05);而HO-1抑制组较AngⅡ组心肌细胞凋亡率及HO-1蛋白表达水平均无明显变化(P>0.05)。结论   HO-1对AngⅡ诱导的乳鼠心肌细胞凋亡具有抑制作用。

Abstract:

Objective   To investigate the role of heme oxygenase-1(HO-1) on the apoptosis of myocardial cells induced by angiotensin Ⅱ. Methods   Neonatal Wistar rats’ myocardial cells were cultured and randomly divided into 4 groups: AngⅡ group, HO-1 inhibitor group, HO-1 induction group and the control group. Western blot was used to detect the HO-1 protein expression. Flow cytometry and Hoechst staining were used to detect the cell apoptosis rate. Results   The apoptosis rate of myocardial cell in AngⅡ group was significantly higher compared with the control group (P<0.05). Compared with AngⅡ group, the apoptosis rate of HO-1 inhibitor group significantly decreased(P<0.05), and the HO-1 protein expression significantly increased(P<0.05). But there were no significant changes of the apoptosis rate and HO-1 protein in HO-1 induction group. Conclusion   HO-1could attenuate the myocardial cells apoptosis induced by AngⅡ.
