ulcerative colitis: expression and clinical significance

摘要:

目的   探讨Toll样受体2、4(TLR2、TLR4)与核因子-κB(NF-κB)在溃疡性结肠炎(UC)黏膜组织中的表达,以及三者在UC发病机制中的作用。方法   采用免疫组化法检测70例UC患者肠黏膜组织TLR2、TLR4与NF-κB的表达水平,并与44例结肠息肉患者的正常黏膜组织对比。结果   TLR2、TLR4和NF-κB在UC组表达率均高于对照组(P值均<0.01);且随内镜分级增高,UC组TLR2、TLR4和NF-κB的表达阳性细胞数均呈逐渐增加趋势(P值均<0.05),三者在活动期和非活动期中的表达也存在统计学差异(P值均<0.01),NF-κB分别与TLR2、TLR4呈显著正相关(r=0.823, P<0.01; r=0.752, P<0.01)。结论   UC组织中TLR2、TLR4和NF-κB表达异常,可能在UC发病机制中起重要作用,检测其变化对疾病活动度的评价具有重要的临床意义。

Abstract:

Objective   To investigate expression of Toll-like receptors 2 and 4(TLR2、TLR4) and nuclear factor-κB(NF-κB) in colonic mucosa of ulcerative colitis. Methods   Immunohistochemistry was performed to examine the expression of TLR2、TLR4 and NF-κB in colonic mucosa of UC (n=70). Normal mucosa of colon polyps was included as a control (n=44). Results   The expression of all three proteins in colonic mucosa of UC was higher than that in control (P<0.01). Cell numbers with positive expression were parallel to the endoscopic classification grades in UC (P values<0.05). Expression of the three proteins in active UC was significantly higher than in unactive UC (P values<0.01). Further, the expression of NF-κB was positively correlated with TLR2 and TLR4(r=0.823, P<0.01;r=0.752, P<0.01, respectively). Conclusion   The elevated expression of TLR2、TLR4 and NF-κB in UC may play a role in UC pathogenesis and, hence, may be of clinical significance.
