大鼠脑出血后血肿周围脑水肿及其与NF-κB 和IL-6表达的关系

Expressions of NF-κB and IL-6 around the hematoma and their correlations
with cerebral edema after intracerebral hemorrhage in rats

摘要:

目的   探讨大鼠脑出血(ICH)后核因子-κB(NF-κB)和白介素-6(IL-6)动态表达情况与血肿周围脑水肿的相互关系。方法   120只SD大鼠随机分为1个假手术组和5个ICH模型组,每组20只。采用右侧脑纹状体内注射新鲜自体不凝血建立大鼠脑出血模型。干/湿重法测定脑组织含水量(BWC),尼氏染色显微镜下观察脑组织病理学变化,免疫荧光检测NF-κB和IL-6蛋白的表达部位,免疫组织化学方法检测NF-κB和IL-6蛋白的表达变化。结果   ICH模型组神经细胞内尼氏体丢失明显,脑组织血肿周围NF-κB和IL-6在脑出血后6h开始表达,随时间延长蛋白表达增加,3d达到高峰。各ICH模型组与假手术组之间有统计学差异(P<0.05);脑出血后NF-κB、IL-6表达量与脑水肿周围含水量呈正相关(P<0.05)。结论   脑出血后NF-κB高表达,说明活化的NF-κB可进一步诱导细胞因子IL-6表达,加重炎症反应, 参与出血性脑水肿的病理过程。

Abstract:

Objective   To investigate the expressions of NF-κB and IL-6 around the hematoma and their correlations with cerebral edema in rat′s brain after experimental intracerebral hemorrhage(ICH). Methods   120 SD rats were randomly divided into 1 sham-operated group and 5 ICH model groups(20 in each group). The intracerebral hemorrhage model was established by nonthrombogenic autologous blood injection in rats. The water content of brain(BWC)-was measured with dry/wet weight method. Histopathological changes were determined by Nissl staining. Locations of NF-κB and IL-6 proteins around the hematoma in cerebral tissues were detected by immunofluorescence. NF-κB and IL-6 expressions around the hematoma in cerebral tissues were detected by immunohistochemistry. Results   Nissl′s bodies of nerve cells in ICH model groups were obviously lost. NF-κB and IL-6 expressions in ICH model groups were significantly higher than those in sham-operated group(P<0.05). NF-κB and IL-6 appeared at 6h, then gradually increased and reached a maximum at 3d. Significant differences existed between ICH model groups and sham-operated group(P<0.05). There were positive correlations between the expressions of NF-κB and IL-6 and the cerebral water content(P<0.05). Conclusion   High expressions of NF-κB in the rats of ICH model groups show that activatedNF-κB can aggravate inflammatory reactions by inducing cytokine IL-6 expression, and take part in the pathological process of brain edema.
