卒中后抑郁大鼠脑内FGF-2表达的研究

摘要:

目的   研究卒中后抑郁大鼠脑内的成纤维细胞生长因子( FGF-2)的表达,探讨FGF-2与卒中后抑郁的关系。方法   采用线栓大脑中动脉(MCAO)法制作卒中SD大鼠模型,将成模后的大鼠随机分为实验组和对照组。实验组采用孤养联合CMS来制作卒中后抑郁组,对照组为卒中大鼠,采用Western印迹法检测实验组与对照组大鼠额叶、海马组织中FGF-2蛋白表达水平,采用实时定量RT-PCR技术,检测两组间大鼠额叶组织中fgf-2 mRNA表达水平。结果   实验组大鼠的额叶组织中FGF-2蛋白和fgf-2mRNA表达水平显著低于对照组(P<0.01),海马组织中FGF-2蛋白表达差异不显著(P>0.05)。结论   卒中后抑郁大鼠额叶FGF-2表达水平下降是引起卒中后抑郁的原因之一。

Abstract:

 Objective   To investigate expression of fibroblast growth factor2(FGF-2) in the brain of post stroke depression(PSD) rats, and the relationship between FGF-2 and PSD. Methods   A rat stroke model was established by means of middle cerebral artery occlusion (MCAO), and then rats were randomly divided into the experimental group and the control group. The experimental group consisted of PSD rats established by means of separation and chronic unpredictable mild stress(CMS), post stroke rats were used as controls. Western blot was used to analyze protein expression of FGF-2 in the frontal lobe and hippocampus tissues. Expression of fgf-2 mRNA was determined in the rat frontal lobe tissue by real-time quantitative PCR. Results   Expression levels of FGF-2 and fgf-2 mRNA in the frontal lobe tissue were significantly lower in the experimental group than in the control group(P<0.01). Protein expression of FGF-2 in the hippocampus showed no significant difference between the two groups(P>0.05) . Conclusion   Decreased expression of FGF-2 in the rat frontal lobe is one of the reasons for PSD.
