黄芪甲苷对大鼠脑缺血再灌注后星形胶质细胞的影响

Effects of astragaloside Ⅳ on the proliferation of astrocytes after cerebral ischemia-re perfusion injury in rats

摘要:

目的     研究黄芪甲苷(AST)对大鼠脑缺血再灌注后星形胶质细胞的影响。方法     将大鼠随机分为假手术组、模型(IR)组、AST 10 、40、100mg/kg组。各干预组分别于缺血前30 min注射相应剂量的AST。采用线栓法制作大鼠一侧大脑中动脉缺血再灌注模型,通过免疫组织化学方法观察各组梗死区和半暗带区胶质纤维酸性蛋白(GFAP)表达水平的变化情况。结果      与假手术组相比,IR组GFAP表达增多(P<0.01);与IR组相比, AST 40、100mg/kg组半暗带区GFAP表达减少(P<0.05);AST各组大鼠缺血中心区GFAP表达变化不大(P>0.05);与AST 10mg/kg组相比,AST 40mg/kg组半暗带区GFAP表达减少(P<0.05)。结论      AST可通过抑制脑缺血再灌注后半暗带区星形胶质细胞增生起到脑保护作用。

Abstract:

Objective      To investigate the effect of astragaloside Ⅳ(AST) on the proliferation of astrocytes after cerebral ischemia-reperfusion(IR) injury in rats. Methods       All SD rats were randomly divided into the sham operation group, the IR group and AST treatment groups (10, 40 and 100mg/kg). Treatment groups were injected with AST 30 minutes before operation. The middle cerebral arterial ischemia/reperfusion (IR) rat model was established by a modified Longa occlusion method. Expression level of glial fibrillary acidic protein(GFAP) was observed in core and penumbral areas by the immunohistochemical technique.  Results     Compared to the sham operation group, expression of GFAP was obviously increased in the IR group (P<0.01). Compared with the IR group, expression of GFAP in the penumbra area diminished in AST 40 and 100 mg/kg groups (P<0.05). Expression level of GFAP in the core area insignificantly changed  (P>0.05). Compared with the AST 10mg/kg group, expression of GFAP in the penumbra area diminished in the AST 40mg/kg group (P<0.05). Conclusion       AST can protect the ischemic brain tissue by inhibiting proliferation of astrocytes in the penumbra area after cerebral ischemia-reperfusion(IR) injury.
