通心络对糖尿病大鼠心肌RAGE、NF-κB的影响

Effects of Tongxinluo on the expressions of RAGE and NF-κB in diabetic rats

摘要:

目的      研究通心络对糖尿病大鼠心肌组织糖基化终末产物受体(RAGE)及核转录因子(NFκB)表达的影响,探讨其对糖尿病大鼠心肌的保护作用及机制。方法      雄性Wistar大鼠60只,随机选出15只作为正常对照组(C组,n=15),剩余45只尾静脉注射0.1%链脲佐菌素(STZ)建立糖尿病模型,成模大鼠40只,随机分为糖尿病模型组(DM 组,n=20) 和糖尿病通心络治疗组(TXL组,n=20),通心络500mg/(kg·d)。喂养24周后,各组大鼠存活数为 C组12只、DM组9只、TXL组13只,处死大鼠迅速心脏穿刺取血,测空腹血糖、糖化血红蛋白(HbA1c) 及糖基化终末产物(AGEs)等指标。RT-PCR及Western blot法测定大鼠心肌组织中RAGE和NFκB的表达。结果     与C组相比,DM组大鼠血清中AGEs水平及心肌组织中RAGE和NF-κB的表达显著升高(P均<0.05)。经通心络治疗后,TXL组大鼠血清中AGEs的水平与DM组无统计学差异(P>0.05),但心肌组织中RAGE和NF-κB的表达较DM组显著降低(P均<0.05)。结论      通心络可能通过抑制糖尿病大鼠心肌组织RAGE及NF-κB的表达而对糖尿病大鼠心肌组织起到一定的保护作用。

Abstract:

Objective     To investigate the effects of Tongxinluo on the expressions of RAGE and NF-κB in diabetic rats. Methods      15 rats were chosen randomly from 60 healthy male Wistar rats as the normal control group(C group, n=15). The other 45 rats were subjected to 0.1% STZ through the tail veins to induce diabetes. Then 40 rats were randomly divided into two groups: the diabetic model group (DM group, n=20) and the Tongxinluo-treatment diabetic group (TXL group, n=20, Tongxinluo 500mg/(kg·d)). After 24 weeks,there were 12 rats in C group,9 ones in DM group and 13 ones in TXL group.All rats were sacrificed, and the fasting plasma glucose, glycosylated hemoglobin and AGEs were determined. Myocardial expressions of RAGE and NF-κB were detected by RT-PCR and Western blot. Results     Compared with C group, Plasma AGEs and myocardial expressions of RAGE and NF-κB were higer in DM group(P<0.05). As for AGEs,there was no significant difference between DM group and TXL group(P>0.05), however, the myocardial expressions of RAGE and NF-κB of TXL group were both lower than those of DM group(P<0.05).  Conclusion     Tongxinluo may exert protective effects on myocardium by suppressing the expressions of RAGE and NF-κB in diabetic rats.
