白细胞介素-17对肝星状细胞增殖及细胞周期的影响

Effect of the interlukin-17 on proliferation and cell cycle of the hepatic stellate cells

摘要:

目的     研究白细胞介素-17(IL-17)对肝星状细胞的增殖及其对细胞周期变化的影响。方法    以体外培养的肝星状细胞为研究对象,分别以200、400、600、800、1000μg/L (终质量浓度)的IL17刺激,并设空白对照组,分别培养24、48、72h。用细胞计数法观察细胞的生长情况,MTT法检测IL-17刺激后肝星状细胞的增殖,流式细胞术检测不同浓度的IL17对肝星状细胞增殖周期的影响。结果    IL-17对细胞的刺激增殖作用在一定的程度和范围内呈现剂量和时间依赖性,200μg/L的IL-17对细胞无明显的刺激作用(P>0.05),800μg/L的IL-17在细胞培养72h后对细胞的刺激增殖作用最明显(P<0.01),1000μg/L的IL-17对细胞则有抑制作用(P<0.01)。IL-17刺激肝星状细胞后能促使细胞由G0/G1期进入S期,细胞增殖以S期细胞比例为主。在一定范围内,细胞增殖指数随着IL-17浓度的增长而增加,而1000μg/L的IL-17则抑制细胞周期,使细胞周期停滞于G1期(P<0.01)。结论    IL-17能刺激肝星状细胞的增殖,在一定的程度和范围内呈现剂量和时间依赖性,IL-17能够促进肝星状细胞由G0/G1期进入S期。

Abstract:

Objective    To investigate the effect of interlukin-17(IL-17) on proliferation and the cell cycle of hepatic stellate cells(HSC). Methods    HSC were treated with IL-17(200, 400, 600, 800, or 1000μg/L)for 24, 48 and 72 hours. HSC in the blank control group were treated without IL-17. The effects of IL-17 on proliferation and the cell cycle were detected by MTT assay and flow cytometry, respectively. Results    The quantity of the cells was associated with the dose of IL-17 and the incubation time. IL-17(200μg/L) had no significant stimulation effect on HSC(P>0.05), while IL-17(800μg/L) for 72h had the most significant stimulation effect (P<0.01). IL-17(1000μg/L) could inhibit the proliferation of HSC(P<0.01). IL-17 could change the distribution of cells at different phases of the cell cycle, promoting the G1 phase cells entering into the S phase. IL-17(1000μg/L) could block cells at the G1 phase(P<0.01).  Conclusions    IL-17 could stimulate the proliferation of HSC, which is associated with the dose of IL-17 and the incubation time, and it could promote G0/G1 phase cells entering into the S phase.
