川续断和丹参影响大鼠正畸牙移动的比较

Effect of Dipsacaceae and Salvia on orthodontic teeth movement

摘要:

目的    研究中药川续断和丹参在大鼠正畸牙移动过程中对其牙周组织改建的作用,并比较两种不同中药在改建过程中的异同。方法    选取72只8周龄SPF级Wistar雌性大鼠,建立大鼠正畸牙移动实验模型,随机分为川续断组、丹参组和对照组3组,每组24只,川续断组每日灌服6g/kg川续断水煎剂,丹参组每日灌服6g/kg丹参水煎剂,对照组每日灌服3mL生理盐水,每隔7d加力1次。3组动物于正畸加力7、14、21、28d分批次处死,每批次6只。剥离头颅骨,分别测量牙移动的距离及牙槽骨的密度,同时制作上颌第一磨牙区牙周组织切片,光学、电子显微镜观察牙周组织改建情况,并进行统计学分析。结果    川续断组、丹参组牙移动距离差异无统计学意义(P>0.05),但是二者均大于对照组,差异有统计学意义(P<0.05);光镜下观察3组破骨细胞数均呈现先增加后平缓趋势,川续断组、丹参组比对照组增加更为显著(P<0.05),川续断组和丹参组之间差异无统计学意义(P>0.05)。3组牙槽骨密度呈现降低趋势,川续断组、丹参组比对照组降低缓慢(P<0.05),川续断组和丹参组之间差异无统计学意义(P>0.05)。结论    灌服丹参和川续断水煎剂均促进大鼠正畸牙移动过程中破骨细胞的增殖和分化,加快牙槽骨吸收及其修复重建,有利于正畸牙移动。

Abstract:

 Objective    To detect and compare the effect of the traditional Chinese drugs, Dipsacaceae and Salvia, on the reconstruction of paradentium in orthodontic teeth movement. Methods    72 female, 8-week-old SPF Wistar rats were selected and the animal models for orthodontic tooth movement were established. The rats were divided into three groups randomly: Dipsacaceae group, Salvia group and control group. Each group had 24 rats. Dipsacus asperoides decoction(6g/kg), Salvia decoction(6 g/kg)  and 3mL NS were drenched daily to the rats in the Dipsacaceae group, Salvia group and  control group respectively.  Stability of the appliances were checked everyday and the applied force was raised by adjusting the springs every 7 days. The rats were executed in batch on the 7th、14th、21st、28th day by peeling off the skulls. Distances of the teeth movement were measured and slices of the periodontium of the maxillary first molar were observed under optical microscope. Data were analyzed with PASWstatistics18. Results    Teeth movement in the Dipsacaceae group and Salvia group were greater than control group(P<0.05)while  no significant difference(P>0.05)was found between them. Light microscope revealed the number of osteoclasts in all 3 groups increased rapidly at the beginning but flatly later. Osteoclasts in  Dipsacaceae and Salvia group showed much more increase than the control (P<0.05), however, no significant difference existed between them(P>0.05). Though alveolar bone density showed the trend of decrease in all 3 groups, Dipsacaceae and Salvia group decreased slowly(P<0.05) and nosignificance was discovered between the two(P>0.05). Conclusion    Salvia and Dipsacaceae decoction can promote the multiplication and differentiation of the osteoclasts in rats, benefit  alveolar bone reconstruction and accelerate orthodontic tooth movement.
