Th17、Treg细胞及其相关细胞因子在子痫前期患者胎盘组织中的表达

摘要:

目的    探讨Th17、CD4+CD25+调节性T(Treg)细胞及其相关细胞因子在子痫前期中的表达及在其发病过程中的作用。方法     选取子痫前期患者胎盘组织30例及正常妊娠胎盘组织(对照组)25例,采用实时荧光定量PCR法检测胎盘组织中Treg细胞转录因子Foxp3和与Th17表达相关的细胞因子孤儿核受体RORc、IL-17及与调节Th17、Treg细胞发育和功能相关的IL-23、IL-27及 IL-27受体mRNA的表达量。结果      子痫前期组胎盘组织中Treg细胞转录因子Foxp3表达较对照组明显降低,差异有统计学意义(P<0.05);子痫前期组胎盘组织中与Th17表达相关的细胞因子RORc、IL17表达与对照组相比明显升高,差异有统计学意义(P<0.05);而与对照组相比,子痫前期组胎盘组织中IL-23、IL-27、IL-27受体表达也明显升高,差异均有统计学意义(P<0.05)。结论     Th17、Treg细胞及其相关细胞因子的表达在子痫前期中均有明显变化,可能参与了子痫前期的发病过程。

Abstract:

Objective      To investigate expressions of Th17, regulatory T-lymphocytes (Tregs) and their related cytokines in pre-eclampsia and the role in the pathogenesis of preeclampsia. Methods    30 placental tissues of patients with pre-eclampsia and 25 placental tissues of women with normal pregnancy (the control group) were selected. Using realtime fluorescent quantitative PCR, mRNA expressions of the special transcription factor of Tregs Foxp3, Th17-related cytokines IL-17 and retinoic acid receptorrelated orphan receptor C(RORc), regulators for the development and function of Th17 and Tregs IL-23 and IL-27, and IL-27R were quantified, in placental tissues of the pre-eclampsia group and the control group. Results     Placenta Foxp3 expression in the pre-eclampsia group was significantly lower than that in the control group(P<0.05); expressions of Th17-related cytokines RORc and IL-17 in the pre-eclampsia group was significantly higher than those in the control group(P<0.05);  also, expressions of IL-23, IL-27 and IL27R in the pre-eclampsia group were obviously increased (P<0.05). Conclusion     Th17, Treg cells and their related cytokines are significantly changed in pre-eclampsia, and may involved in the pathogenesis of pre-eclampsia.
