ADAM33基因F+1 位点单核苷酸多态性与支气管哮喘的相关性研究

Association between the ADAM33 gene polymorphism and bronchial asthma

摘要:

目的    检测裂解素-金属蛋白酶33基因 (ADAM33)F+1位点单核甘酸多态性在支气管哮喘患者中的分布频率, 并探讨其与支气管哮喘的相关性。 方法     采用等位基因特异性聚合酶链反应(ASPCR)技术及 DNA 测序的方法, 对哮喘组 126 例患者及对照组 121 例健康者进行 ADAM33 基因 F+1 位点单核苷酸多态性分析。结果     ADAM 33基因F+1位点哮喘组和对照组基因型分布均符合HardyWeinburg定律;ADAM33 基因 F+1 位点 3 种基因型(CC、CT、TT)在哮喘组分布分别为 68(54.0%)、48( 38.1%)、10( 7.9%),在对照组分布分别为 70(57.9%)、46(38.0%)、5( 4.1%),无显著性差异(χ2=1.638, P>0.05)。哮喘组和对照组 ADAM33 基因 F+1 位点等位基因C和T的频率分别为 0.730、 0.270、0.769、 0.231,两组等位基因频率比较差异无显著性 (χ2 =0.970,P>0.05)。结论     ADAM33 基因 F+1 位点在支气管哮喘患者中存在CC、CT、TT 多态性,ADAM33 基因 F+1 位点基因多态性与支气管哮喘无明显相关性。

Abstract:

Objective    To investigate the association between polymorphism of the F+1 locus allele in the ADAM33 gene and bronchial asthma. Methods    Allele-specific polymerase chain reaction (AS-PCR) and DNA sequencing were used to determine polymorphism of the F+1 locus allele in the ADAM33 gene in 126 unrelated patients with bronchial asthma and 121 unrelated healthy controls. Results     Genotype distributions of the F+1 locus allele in the ADAM33 gene in the patient group and the control group did not deviate from the HardyWeinburg equilibrium. Frequencies of the genotypes (CC,CT,TT) were 54.0% (n=68), 38.1% (n=48) and 7.9% (n=10) in the 126 asthmatic patients and 57.9% (n=70), 38.0% (n=46) and 4.1% (n=5) in the 121 controls, respectively. There was no significant difference in distribution of the genotypes (CC,  CT, TT) between asthmatic patients and healthy controls (χ2=1.638, P>0.05). Frequencies of the alleles (C, T) were 0.730 and 0.270 in asthmatic patients and 0.769 and 0.231 in the controls, respectively, and there was no significant difference in allele frequency between them (χ2 =0.970,P>0.05). Conclusion      Polymorphism of the F+1 locus allele in theADAM33 gene(CC,CT,TT) exists in asthmatic patients, but the polymorphism is not significantly associated with the susceptibility to bronchial asthma.
