脑神康胶囊对高糖环境下氧化损伤大鼠海马神经元HIF-1α表达的影响

Effect of the Naoshenkang capsule on expression of hypoxia-inducible factor-1 alpha  of oxidative damaged rat hippocampal neurons in high glucose

摘要:

目的   研究高糖环境下体外培养大鼠海马神经元氧化损伤时乏氧诱导因子-lα(HIF-lα)表达的变化以及脑神康胶囊对神经元的保护作用。方法  原代培养大鼠海马神经元,将细胞随机分成8组:对照组、高糖组、黄嘌呤/黄嘌呤氧化酶组(X/XO组)、川芎嗪组、生理盐水组、脑神康低(5mL/kg)、中(10mL/kg)、高浓度组(20mL/kg),对照组用低糖型DMEM培养基培养,其余7组用葡萄糖浓度为25mmol/L的DMEM培养基培养。除对照组及高糖组正常培养,其余各组均用黄嘌呤/黄嘌呤氧化酶处理建立氧化损伤模型,其中中药各浓度组及川芎嗪组分别于加入黄嘌呤/黄嘌呤氧化酶之前换以不同浓度含药血清和川芎嗪的DMEM培养液,采用Real-Time PCR法检测各组海马神经元HIF-1α mRNA的表达。结果  与对照组相比, 高糖组及X/XO组HIF-lα mRNA表达均明显上升(P均<0.01); 与高糖组相比,X/XO组HIF-1α mRNA的表达量升高(P<0.05);与X/XO组比较,川芎嗪组及中药各浓度组HIF-1α mRNA的表达量均降低,其中中浓度组最低,具有明显的统计学意义(P均<0.01),川芎嗪组与低浓度组接近,无统计学差异(P>0.05)。〖结论  脑神康胶囊对高糖环境下培养的大鼠海马神经元氧化损伤有明显的保护作用,其作用机制可能与其抑制HIF-1α的表达、改善组织缺氧密切相关。

Abstract:

Objective  To research the effect of the Naoshenkang capsule on expressionof hypoxia-inducible factor-1 alpha of oxidative damaged rat hippocampal neuronsin high glucose. [JP2]Methods  The primarily cultured neonate rat hippocampal neurons were randomly divided into eight groups: the control group, the high glucose group, the xanthine/xanthine oxidase group(X/XO group), the ligustrazine group, the normal sodium group and the small, middle, and large dosage of Naoshenkang groups. The control group was raised in 5.5mmol/L DMEM, and the other groups treated by xanthine/xanthine oxidase except that the high glucose group was raised in 25mmol/L DMEM. Themethod of Real-Time PCR was used to detect mRNA expression of HIF-1α. Results  Expression of HIF-lα mRNA of the high glucose group was significantly higher than that of the control group (P<0.01), but lower than that of the X/XO group (P<0.05). Expression of HIF-lα mRNA of the ligustrazine group and all Chinese Medicine groups were significantly lower than that of the X/XO group, and the middle dosage of Chinese Medicine groups was the lowest (P<0.01).Conclusions  High glucose can cause injury of hippocampal neurons and the Naoshenkang capsule has an inhibitory effect on it. Its mechanism may be that the Chinese Medicine can inhibit expression of HIF-lα mRNA and improve tissue hypoxia.
