完全脾切除阻止孕期小鼠胰岛β细胞总量增加

Total splenectomy prevents the increase of islet beta cells in pregnant mice

摘要:

目的  探讨脾对小鼠怀孕状态下胰岛β细胞增加的影响。方法  40只7周龄昆明雌性小鼠分为4组:脾切除怀孕组、假手术怀孕组、未受孕脾切除对照组和假手术对照组,每组10只。不同时间收集血液,分析小鼠空腹血糖。小鼠分娩后,立即切取胰腺组织,进行免疫组化染色,用图像分析系统分析胰岛β细胞总重量。分离纯化小鼠胰岛及脾细胞,在Transwell小室进行非接触式共培养,CCK-8处理后,酶标仪490?nm处比色,分析胰岛细胞增殖状况。结果  与假手术对照组(7.03±2.71 )mg相比,假手术怀孕组胰岛β细胞总量(11.89±2.83)mg显著增加(P<0.05);脾切除怀孕组胰岛β细胞总量(6.10±2.12)mg低于假手术怀孕组(11.89±2.83)mg,P<0.05,但与脾切除对照组(5.37±2.11?mg)比较差异无统计学意义(P>0.05)。假手术怀孕组和脾切除怀孕组空腹血糖均在正常范围。体外细胞共培养研究结果表明,脾细胞可促进胰岛β细胞增殖。结论  脾可促进小鼠孕期胰岛β细胞总量的增加,其机制可能涉及脾分泌的某些因子的间接作用。脾切除可限制孕期小鼠胰岛β细胞总量的增加。

Abstract:

Objective  To explore whether the spleen can affect the increase of isletβ cell mass in pregnant mice. Methods  40 female KM mice, aged7 weeks, were divided into four groups: the splenectomized pregnant group, the sham pregnant group, the splenectomized control group and the sham control group without pregnancy. Fasting bloodglucose was measured in different study periods. After parturition, the pancreas tissue was obtained from various groups of mice and immunohistochemically stained for  β cell mass measurement using an image analysis system. Islet and spleen cells isolated from mice were purified and co-cultured in Transwell. The proliferation of islet cells was measured following CCK-8treatment by an enzyme-labeling instrument at 490?nm. Results  There was a significant increase of islet β cell mass in the sham pregnant group compared with that in the sham control group(11.89mg±2.83mg vs 7.03mg±2.71mg, P<0.05). However, the islet β cell mass in the splenectomized pregnant group was only 6.10mg±2.12mg,which was significantly lower than that in the sham pregnant group(P<0.05). In the splenectomized pregnant group and the sham pregnant group, fasting blood glucose concentration was normal. In vitro cell co-culture showed that spleen cells promotedthe proliferation of islet β cells. Conclusion  Spleen cells canpromote the increase of islet cell mass by secreting some factors; splenectomy may prevent the increase of islet cell mass in pregnant mice.
