姜黄素对小鼠实验性急性坏死性胰腺炎的抑制作用

Curcumin protects against acute necrotizing pancreatitis damage 
in mice by inhibiting NFκB

摘要:

目的探讨姜黄素对急性坏死性胰腺炎小鼠的保护作用及其机制。方法随机将40只健康的KM小鼠分为对照组、急性坏死性胰腺炎(ANP)模型组和低、高剂量姜黄素处理组各10只。在造模成功后12?h采集血样并处死小鼠,取胰腺组织观察病理损伤情况并进行病理学评分,分别采用全自动生化分析仪、ELISA双夹心技术、RTPCR法、Western blot法检测血清淀粉酶活性、血浆TNFα水平、各组胰腺组织NFκB mRNA及NFκB p65总蛋白和胞核蛋白的表达。结果血清淀粉酶、TNFα水平,胰腺组织病理学评分及NFκB表达ANP组较对照组明显增高(P<0.05),低、高剂量姜黄素治疗组较ANP组显著降低(P<0.05)。结论姜黄素可有效减轻急性坏死性胰腺炎小鼠胰腺损伤,其机制可能与降低NFκB活性有关。

Abstract:

To investigate the protective effect of curcumin on the development of acute necrotizing pancreatitis in mice. MethodsKunming mice were randomly divided into the control group, ANP group, low concentration of curcumin group, and high concentration of curcumin group. At 12h after induction of ANP, a blood sample was withdrawn for analysis of amylase activity and TNFα level, the serum amylase was determined by an Automatic Biochemical Analyzer, and expression of TNFα was determined by ELISA. Pancreatic pathological damages were evaluated to observe the pathological score. Expression of NFκB was determined by RTPCR and Western blot methods. Results  The serum amylase, TNFα level, pathological scores and expression of NFκB in the ANP group were significantly higher than the in control group(P<0.05), but they were significantly lower in the two roglitazone curcumin groups than in the ANP group(P<0.05). ConclusionCurcumin may effectively alleviate pancreas injury in acute necrotizing pancreatitis in mice by inhibition of NFκB expression.
