乳腺癌转移相关蛋白蛋白质组学初步研究

Metastases associated with proteins in breast cancer by proteomics

摘要:

目的筛选人乳腺癌腋窝淋巴结转移相关蛋白,并对所筛选的两个差异蛋白进行验证。方法选取有淋巴结转移和无淋巴结转移的乳腺癌新鲜标本各10例,提取组织蛋白进行双向凝胶电泳(2DPAGE),通过对比分析筛选出差异蛋白点并进行质谱鉴定(MALDITOFMS)。免疫组织化学SP法检测72例乳腺癌组织中差异表达蛋白Stathmin和Ezrin。结果质谱鉴定出214个蛋白位点。排除胶原蛋白、血清白蛋白及电泳弥散等造成的部分相同蛋白后,筛选出表达差异≥3.0倍的蛋白36个。免疫组织化学检测显示Stathmin、Ezrin蛋白在乳腺癌淋巴结转移组和无淋巴结转移组阳性率分别为73.5%、47.4% 和78.6%、46.9%,两组间差异具有统计学意义(P<0.05)。结论淋巴结转移组和无转移组乳腺癌组织存在多种差异蛋白。Stathmin、Ezrin蛋白可能参与了乳腺癌的浸润与转移过程。

Abstract:

To screen the associated proteins of breast cancer which have axillary lymph node metastases. Methods20 specimens of fresh breast cancer tissues were collected and classified into two groups with and without lymph node metastases, with 10 cases in each group. Proteins were extracted from these specimens and separated by 2DPAGE. Distinct protein spots were selected for mass spectrometry. Stathmin and Ezrin protein expression was assayed by an immunohistochemical SP method in 72 breast cancer cases. Results214 protein spots were identified.36 proteins with at least 3.0fold of differential expression were identified after collagen protein, serum albumin and identical proteins which were caused by electrophoresis dispersion were eliminated. Immunohistochemistry showed that the positive rate of Stathmin and Ezrin proteins was 73.5%, 47.4%, 78.6%, and 46.9% in breast cancer with and without lymph node metastases. Differences of the two sets had statistical significance (P<0.05). ConclusionThere are many distinct proteins between breast cancer with and without lymph node metastases. Stathmin and Ezrin may be involved in the process of progression and metastasis of breast cancer.
