改良Hemistraw与Cryoleaf法对人卵和胚胎玻璃化保存效果的比较

Vitrification of human oocytes and embryos:modified Hemistraw versus Cryoleaf

摘要:

目的比较改良的Hemistraw与Cryoleaf法对人卵子和胚胎的玻璃化保存效果。方法 将接受体外受精-胚胎移植周期中剩余的98枚未受精卵和76枚废弃胚胎,分别采用Cryoleaf(A组)与改良的Hemistraw(B组)为载体进行玻璃化冷冻,复苏后观察并计算两种玻璃化载体的卵子存活率,24?h继续存活率与卵裂率;胚胎存活率,24?h继续存活率及囊胚形成率。结果Cryoleaf与改良的Hemistraw法保存人未受精卵分别为43、55枚,复苏存活分别为29(67.4%)、31(56.4%)枚,24?h继续存活分别为23(53.5%)、25(45.5%)枚,卵裂分别为3(7.0%)、1(1.8%)枚,两种载体差异不明显(P>0.05)。Cryoleaf与改良的Hemistraw法保存废弃胚胎分别为39、37枚,复苏存活分别为29(74.4%)、33(89.2%)枚,24?h继续存活分别为28(71.8%)、33(89.2%)枚,形成囊胚分别为5(12.8%)、1(2.7%)枚,两种载体差异不明显(P>0.05)。结

Abstract:

To evaluate the results of vitrified human unfertilized oocytes and discarded embryos by selfmade modified Hemistraw or Cryoleaf. Methods98 unfertilized oocytes and 76 discarded embryos, which derived from conventional IVF or ICSI cycles, were randomly divided into group A(Cryoleaf)and B(modified Hemistraw). After thawing, the frozen samples were continuously observed. The survival rate of unfertilized oocytes, the 24?h survival rate  and the cleavaged rate were measured. Also the survival rate of embryos, the keeping survival rate of 24?h and the blastocyst rate were calculated. ResultsIn group A, 43 unfertilized oocytes were vitrified, of them 29 (67.4%) survived, 23(53.5%) kept survival for 24?h, and 3(7.0%) were cleavaged. In group B, 55 unfertilized oocytes were vitrified, of them 31(56.4%) survived, 25 (45.5%) survived for 24?h, and 1 (1.8%) had cleavage. The above indexes had no statistical significances between the two groups(P>0.05). 39 and 37 discarded embryos were respectively vitrified by Cryoleaf and modified Hemistraw, the survival rate of embryos, the 24?h survival rate and the blastocyst rate showed no significant differences (74.4% vs 89.2%, 71.8% vs 89.2%, and 12.8% vs 2.7%, P>0.05).ConclusionThe modified Hemistraw may be an alternative  method to Cryoleaf due to its widespread resource, low price and similar survival rate of vitrification.
