侧脑室注射内吗啡肽-1降低神经病理性疼痛大鼠海马和PAG IL-6mRNA的表达

摘要: 目的:观察侧脑室微量注射内吗啡肽1(endomorphin1, EM1)对坐骨神经慢性压迫性损伤(chronic constriction injury, CCI)大鼠痛阈以及海马和中脑导水管周围灰质(periaqueductal gray,PAG)中白细胞介素6(interleukin6, IL6)mRNA表达的影响。方法:建立大鼠慢性坐骨神经压迫性损伤模型,侧脑室微量注射EM1(20?μg/20?μl),手术前1?d和手术后第7d观察其机械性痛敏和热痛敏的变化,术后第7?d将大鼠处死,采用原位杂交观察海马和PAG IL6 mRNA的表达,假手术及侧脑室注射生理盐水作对照。结果:实验发现CCI手术导致大鼠机械性痛阈和热痛阈明显下降,且增加大鼠海马和PAG部位IL6mRNA的表达。侧脑室注射EM1可提高大鼠基础痛阈,且显著改善大鼠神经病理性疼痛状态,降低海马和PAG部位IL6mRNA的表达。结论:侧脑室微量注射EM1可有效抑制神经病理性疼痛,且明显降低海马和PAG IL6的表达。

Abstract: To investigate the effect of intracerebroventricular (i.c.v.) injection of endomorphin1 (EM1) on IL6 mRNA expression level in hippocampus and on PAG in chronic constriction injury (CCI) model of rats. Methods: The CCI model was established and given endomorphin1 (20 μg/20 μl, i.c.v) once daily for 3 consecutive days. The control rats were operated on and injected NS. The mechanical and the thermal pain threshold were measured 1 day before operation and 7 days after operation. The expression level of IL6 mRNA was measured by in situ hybridization. Results: Mechanical allodynia and thermal hyperalgesia were induced in CCI rats. In addition, the expression level of IL6 mRNA in hippocampus and PAG markedly increased. Intracerebroventricular injection of EM1 increased the basic pain threshold of the rats, reversed the CCIinduced existing neuropathic pain state and significantly suppressed the increased expression level of IL6 mRNA induced by CCI.
