外泌体lncRNA OGFRP1在84例非小细胞肺癌中的表达及临床意义

Expression and clinical significance of exosomal long non-coding RNA OGFRP1 in 84 cases of non-small cell lung cancer

摘要: 目的 探讨长链非编码 RNA(lncRNA)OGFRP1 在非小细胞肺癌(NSCLC)血清外泌体中的表达及临床意义。 方法 分别对84例初诊为NSCLC的患者及84例正常健康者的血清进行外泌体提取鉴定、RNA提取及检测lncRNA表达。统计学分析血清外泌体lncRNA OGFRP1诊断NSCLC的灵敏度、特异度与准确性,分析 lncRNA OGFRP1 的表达与临床病理特征的关系。 结果 84例初诊为NSCLC的患者均经病理学证实为NSCLC。lncRNA OGFRP1 在NSCLC血清外泌体中显著高表达(χ2=96.459,P<0.001)。以病理诊断为金标准,lncRNA OGFRP1测值对NSCLC的辅助诊断的灵敏度、特异度和准确性分别为90.5%、86.9%和93.9%,高于癌胚抗原,差异有统计学意义(P<0.001)。血清外泌体lncRNA OGFRP1的相对表达量与NSCLC癌患者的淋巴结转移(P=0.034)以及T分期(P=0.030)差异有统计学意义。 结论 血清外泌体lncRNA OGFRP1 的检测对于诊断NSCLC具有较高的灵敏度、特异度和准确性,可作为NSCLC早期筛选、检测和分期的新型肿瘤标志物。

Abstract: Objective To explore the expression and clinical significance of long non-coding RNA(lncRNA)OGFRP1 in serum exosomes of non-small cell lung cancer(NSCLC). Methods A total of 84 cases preliminarily diagnosed as NSCLC and 84 healthy controls were enrolled. The serum exosomes were extracted and identified, the RNA was extracted, and lncRNA expression was detected. The sensitivity, specificity and diagnostic accuracy of lncRNA OGFRP1 were statistically analyzed. The relationship between the lncRNA OGFRP1 expression and clinicopathological factors were analyzed. Results All cases were pathologically confirmed as NSCLC. lncRNA OGFRP1 was highly expressed in NSCLC(χ2=96.459, P<0.001). With pathological diagnosis as the gold standard, the sensitivity, specificity and accuracy of lncRNA OGFRP1 in the auxiliary diagnosis of NSCLC were 89.4%, 87.9% and 89.4%, respectively, higher than those of carcino-embryonic antigen(P<0.01). The relative expression of lncRNA OGFRP1 in serum exosomes was significantly associated with lymph node metastasis(P=0.034)and T stage(P=0.030)in NSCLC patients. Conclusion The detection of exosomal lncRNA OGFRP1 has high sensitivity, specificity and accuracy in the diagnosis of NSCLC, which can be used as a novel tumor marker for early screening, diagnosis and staging of NSCLC.
