肿瘤相关巨噬细胞对肝癌细胞c-Met分子表达的影响

Effect of tumor associated macrophages on the expression of c-Met in hepatocellular carcinoma cells

摘要: 目的 探究肿瘤相关巨噬细胞促进肝癌细胞转移的潜在分子机制。 方法 巨噬细胞经刺激后与肝癌细胞共培养,利用RT-PCR和流式细胞术检测肿瘤相关巨噬细胞对肝癌细胞c-Met分子表达的影响,利用Transwell细胞迁移实验检测HGF-c-Met通路对肝癌细胞迁移能力的影响。 结果 THP-1来源的巨噬细胞与肝癌细胞共培养后,肝癌细胞c-Met分子在mRNA和蛋白水平的表达均有增强;与巨噬细胞共培养的肝癌细胞,在重组蛋白HGF诱导作用下,其迁移能力明显增强。 结论 肿瘤相关巨细胞可以上调肝癌细胞c-Met分子的表达,高表达的c-Met分子增强了肝癌细胞的迁移能力。

Abstract: Objective To explore the molecular mechanism of c-Met expression in hepatocellular carcinoma cells(HCC)influenced by tumor associated macrophages. Methods HCC cells were cocultured with macrophages derived from THP-1 cells. The expression of c-Met in HCC cells was detected at mRNA level by RT-PCR and protein level by flow cytometry. The migration of HCC cells was detected by Transwell cell migration test. Results The expression of c-Met in HCC cells was upregulated by the macrophages at both mRNA and protein levels. After HCC cells were cocultured with macrophages, the migration was enhanced in the presence of human recombinant protein hepatocyte growth factor(HGF). Conclusion Tumor associated macrophages promote the migration of HCC cells by upregulating the expression of c-Met in HCC cells.
