金银花提取物对化合物48/80、组胺及P物质诱导小鼠瘙痒模型的作用

Effects of Flos Lonicerae Japonicae extract on pruritus induced by compound 48/80, histamine and substance P in ICR mice

摘要: 目的 探讨金银花提取物对化合物48/80、组胺及P物质诱导的小鼠瘙痒模型的作用。方法 采用0.3、0.8、2.4 g/kg 3个不同剂量的金银花提取物将小鼠灌胃给药1 h后, 分别皮内注射致痒物质化合物48/80、组胺和P物质, 随后将小鼠置于观察笼内, 用摄像仪记录1 h内小鼠的行为。结果 金银花提取物可呈剂量依赖性的抑制P物质诱导的小鼠瘙痒, 而对化合物48/80和组胺诱导的小鼠瘙痒没有明显的抑制作用。结论 金银花提取物对P物质诱导的小鼠模型有明显的抗瘙痒作用, 其抗瘙痒活性主要作用部位为角质形成细胞, 而非肥大细胞。

Abstract: Objective To study the effect of Flos Lonicerae Japonicae on pruritus induced by various pruritic agents including compound 48/80, histamine and substance P in mice. Methods Flos Lonicerae Japonicae extracts(0.3 g/kg, 0.8 g/kg and 2.4 g/kg) were respectively administered orally to ICR mice 1 h before the injection of compound 48/80, histamine and substance P. Immediately after the injection of pruritogens, the animals were put into the acrylic cage again, and their behaviors were videotaped. Results Flos Lonicerae Japonicae dose-dependently inhibited substance P-induced scratching. However, no inhibitory effect was observed on the scratching behavior induced by histamine or compound 48/80. Conclusion Flos Lonicerae Japonicae shows significant antipruritic effect on substance P-induced pruritus in ICR mice. The primary site of its antipruritic activity is epidermal keratinocytes rather than mast cells.
