济南市西部地区鼠类携带路氏锥虫的分子生物学鉴定与流行特征

Molecular identification and prevalence of Trypanosoma lewisi in rodents in western areas of Jinan City

摘要: 目的 探讨济南市西部地区鼠类中携带锥虫的分子流行特征,为锥虫病的传播风险及防治提供依据。 方法 在济南市西部地区采集鼠类脏器样本,应用属通用引物,采用聚合酶链式反应(polymerase chain reaction, PCR)扩增锥虫18S小亚基核糖体RNA编码基因(18S small subunit ribosomal RNA gene, 18S rDNA),对毛细管电泳检测阳性的扩增产物进行测序。对阳性样本进行18S rDNA的序列比对和遗传变异分析。 结果 2020年在济南市西部地区采集鼠100只,对其脏器混合样本进行锥虫18S rDNA扩增,共获得5份锥虫阳性样本,锥虫阳性率为5.00%。5份18S rDNA测序分析结果表明,鼠中携带的锥虫均为路氏锥虫(Trypanosoma lewisi),序列一致性最高为100.00%。 结论 本次调查的济南西部地区鼠类携带的锥虫均为路氏锥虫,为该地区锥虫病的后续比较研究提供了基础数据。

Abstract: Objective To investigate the molecular epidemiological characteristics of Trypanosoma in rodents in western areas of Jinan City and provide evidence for assessing the transmission risk and prevention of trypanosomiasis. Methods Rodent organ samples were collected from western areas of Jinan City. Genus-specific primers were used to amplify the 18S small subunit ribosomal RNA gene(18S rDNA)of Trypanosoma via polymerase chain reaction(PCR). Positive amplification products identified by capillary electrophoresis were sequenced. Sequence alignment and genetic variation analysis of 18S rDNA from positive samples were performed. Results In 2020, a total of 100 rodents were captured from western Jinan. PCR amplification of 18S rDNA from mixed organ samples yielded 5 positive samples, with a Trypanosoma positivity rate of 5.00%. Sequencing and analysis of the five positive 18S rDNA sequences revealed that all detected trypanosomes were Trypanosoma lewisi, with the highest sequence identity reaching 100.00%. Conclusion The trypanosomes carried by rodents in the western areas of Jinan City were exclusively Trypanosoma lewisi, which provides basic data for subsequent comparative studies on trypanosomiasis in this region.
