Determination of curcumol in animal plasma and tissues by HPLC-MS/MS

Abstract:

Objective〓〖WTBZ〗To develop high performance liquid chromatography coupled with the tandem mass spectrometry(HPLC-MS/MS) quantitative detection method for determination of curcumol in animal plasma and tissues. 〖WTHZ〗Methods〓〖WTBZ〗Dog plasma and rat tissue samples spiked with ornidazole as the internal standard were separated on a Diamonsil C18 column and eluted at a flow rate of 1mL/min, and the precursor/ product ion combinations of m/z 254.3→219.4 and m/z 220.3→128.1 were monitored to quantify curcumol by MRM. 〖WTHZ〗Results〓〖WTBZ〗A good linearity of plasma or tissue curcumol concentration and area ratio of curcumol to i.s was obtained. 〖WTHZ〗Conclusion〓〖WTBZ〗The developed method is rapid, highly sensitive and specific and could be used in animal curcumol pharmacokinetic and tissue distribution studies.
