Effects of human amniotic homogenate on blebs and expressions of TGFβ1 and CTGF after experimental trabeculectomies in rabbits

Abstract: To investigate the effects of human amniotic homogenate on blebs and the expressions of TGFβ1 and CTGF following trabeculectomy in rabbits. MethodsThirty New Zealand white rabbits were randomly divided into two groups: the control group and the experimental group. The right eyes were subjected to standard trabeculectomies. After the operations, human amniotic homogenate was topically used in the experimental group four times daily for 14 days and was not used in the control group. The clinical features and the pathological changes were observed on different days after surgeries. Results14 eyes in the experimental group had functional blebs and 8 in the control group(P<0.05). The intraocular pressure of the control group was higher than that of the experimental group after 2 days(P<0.05). The fibroblasts in the experimental group were less than those in the control group(P<0.05). The expressions of TGFβ1 and CTGF were lower in the experimental group than those in the control group(P<0.05). Conclusions The supernatant of a homogenized amnion has no side effects, and it can inhibit the expressions of TGFβ1 and CTGF and improves the filtering function and decreases the intraocular pressure.
