Preparation and characterization of acellular dermal matrix

Abstract:

 Objective   To prepare a new type of acellular dermal matrix (ADM) and analyze its properties. Methods   A new type of ADM was prepared by means of combined treatments with physical, chemical and biological methods, and its structure, composition and properties were evaluated. Results   No cell was observed in ADM, and the content of collagen was about 92.01%. The obtained ADM framework had interconnected pores of about 100μm in diameter, and the natural framework of the collagen kept well. Its water vapor permeability was about 2923g/m2 per day, and the porosity reached 85.54%.  MTT assay indicated that cells cultured on ADM adhered to the surface, proliferated well and were evenly distributed. Conclusion   The prepared ADM has good microstructure, high mechanical properties and porosity, controlled enzymatic stability and good cell compatibility, and it meets the requirement of tissue engineering for stents.
