Polyamine Metabolism and Cancer Therapy

Abstract:

 Metabolism of Polyamine plays an important role in tumor growth and progress. Ornithine Decarboxylase (ODC) and S-adenosylmethionine Decarboxylase (SAMDC) are two key enzymes in the pathway of polyamine synthesis. The antitumor activities of ODC and SAMDC inhibitors have been well-established in in-vitro and animal experiments. However, their applications are limited in clinic due to their toxicity. Recent research found that expression of ODC and SAMDC are up-regulated in tumor tissue and knockdown of these two genes could inhibit tumor cell proliferation and migration. These studies provide new therapeutic approach for cancer prevention and treatment on the targets of polyamine metabolism. In this review, we summarize the new progress on relationship between polyamine metabolism and cancer therapy.
