Yishen Huoxue decoction regulates FN and COL Ⅳ expressions
through the P38MAPK pathway in mesangial cells

Abstract:

To investigate the effects of the Yishen Huoxue decoction on secretion of fibronectin(FN) and collgen Ⅳ(COL Ⅳ) through p38 mitogenactivated protein kinase (p38MAPK) in rat mesangial cells. MethodsPharmacologic serum was prepared by the serum pharmacologic method. The cultured mesangial cells in vitro were divided into three groups: the control group(the normal rat blood serum), the stimulate group (the normal rat blood serum+10%IL1β), and the decoction group(traditional Chinese drug +10%IL1β). Levels of FN and COL Ⅳ were determined by ELISA. Changes of phosphated p38MAPK protein andαSMA were determined by Western blot analysis. ResultsCompared with the stimulate group, FN and pp38MAPK expressions of the decoction group significantly decreased(P<0.01), and expressions ofαSMA and COLⅣ were  also inhibited(P<0.01). ConclusionYishen Huoxue decoction can decrease expression of αSMA and alleviate levels of FN and COLⅣ, and the p38MAPK pathway may be involved in the mechanism.
