Cancer initiating cells of lung cancer cell line A549

Abstract: To investigate the fraction of cancer initiating cells in lung cancer cell line A549 by cloning and tumorigenic analyses. MethodsA clone formation assay was carried out to determine the clonogenicity and regeneration ability of cells. Cells from a clone were injected into BALB/Cnude mice to test tumorigenicity. To determine whether CD133- cells of A549 possess the characteristics of cancer stem cells, we isolated the cells by magnetic separation and tested whether Hoechst 33342 staining could affect clonogenicity of the A549 cells by clone formation assay. ResultsMore than 45% of the A549 cells formed large clones that were able to generate subclones and subsequently give rise to xenograft tumors in the BALB/Cnude mice. Both CD133+ cells and CD133- cells of A549 possessed stem cell characteristics, and Hoechst 33342 affected the clonogenicity and proliferation of a single A549 cell. ConclusionMore than 45% of A549 cells are cancerinitiating cells.
