Effects of cholecystokinin-octopeptide on endoplasmic reticulum stress induced by LPS in mouse monocyte RAW264.7 cells

Abstract: Objective To investigate the effects of cholecystokinin-octopeptide (CCK-8) on ER stress (ERS) induced by lipopolysaccharide (LPS) in mouse monocyte RAW264.7 cells. Methods RAW264.7 cells were divided into control group, LPS group, CCK-8 grpup, devazepide group(DEV group), CCK-8+LPS group, and DEV+CCK-8+LPS group. The active form of X-box binding protein 1(XBP1s) mRNA expression was assayed by RT-PCR, glucose regulating protein 78 (GRP78) and C/EBP homologous protein (CHOP) protein expressions were determined by Western blotting, and IL-1β expression levels were measured by ELISA. Results Compared with control group, the expressions of XBP1s mRNA and GRP78 and CHOP protein in LPS group increased (P<0.05); cmpared with LPS group, the expressions of XBP1s mRNA and GRP78 and CHOP protein in CCK-8+LPS group decreased (P<0.05); compared with CCK-8+LPS group, the expressions of XBP1s mRNA and GRP78 and CHOP protein in DEV+CCK-8+LPS group increased (P<0.05). The changes of IL-1β contents showed the same trend as that of CHOP expression (P<0.05). Conclusion CCK-8 exerts an anti-inflammatory effect by inhibiting LPS-induced ERS and decreasing the expression of IL-1β in RAW264.7 cells.
