“促愈瘢痕消”对兔耳增生性瘢痕作用的实验研究

The effect of Cuyubanhenxiao on hypertrophic scars in rabbit ears

摘要: 目的:探讨“促愈瘢痕消”对兔耳增生性瘢痕的作用。方法:用新西兰白兔12只,均在右侧兔耳腹侧面制作增生性瘢痕动物模型。待瘢痕形成后随机分为两组,每组6只:试验组(A组)外用“促愈瘢痕消”,对照组(B组)外用生理盐水,均1次/d,连用1月。分别在瘢痕形成时、治疗后4周测量瘢痕厚度,治疗后1、4周切取瘢痕组织做组织病理学及p53、增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)测定。结果:A组在4周时较B组瘢痕厚度明显变薄,颜色变浅,质地变软,胶原色淡而排列稀疏,随用药时间延长差异明显;两组间瘢痕厚度差异有统计学意义(P<0.05);p53、PCNA阳性表达率在1、4周时两组间差异有统计学意义(P<0.05),A组在1、4周时差异有统计学意义(P<0.05),B组在1、4周时差异无统计学意义(P>0.05)。结论:“促愈瘢痕消”能够明显抑制兔耳瘢痕的增生,且机理可能是减少p53及PCNA的表达,有望成为治疗增生性瘢痕的有效药物。

Abstract: Objective: To explore the effect of Cuyubanhenxiao on hypertrophic scars in rabbit ears. Methods: Twelve adult New Zealand white rabbits were given hypertrophic scars over the ventral surface of the right ears and were randomly divided into the Cuyubanhenxiao group (group A ) and the control group (group B). They were subjected to the drugs once every day for 1 month. The thickness of the scars was determined at the formation and at 4 weeks after treatment. Immunohistochemical (P53 and PCNA) and histomorphology were used to determine the scar tissues at 1 and 4 weeks. Results: The thickness of the scar was significantly different between the two groups (P<0.05). Expression of p53 and PCNA was significantly different between the 1 week and 4 weeks groups (P<0.05). Expression of p53 and PCNA was significantly different in group A between the 1 week and 4 weeks groups (P<0.05), but there was no significant difference in group B between the 1 week and 4 weeks groups(P>0.05). Conclusions: Cuyubanhenxiao can heal hypertrophic scars of rabbit ears, which may be caused by the decrease of the expression of p53 and PCNA.
