祛瘀消斑胶囊和辛伐他汀对动脉粥样硬化生长因子和MMP1 mRNA表达的影响

摘要: 目的:观察祛淤消斑胶囊和辛伐他汀对动脉粥样硬化生长因子和基质金属蛋白酶1(collagen I matrix metalloproteinase 1,MMP1)及其组织抑制物1(tissue inhibitor 1,TIMP1)表达的影响。方法:对实验性动脉粥样硬化兔,分别给予辛伐他汀和中药祛淤消斑胶囊治疗12周,应用免疫组化和逆转录多聚酶链反应观察动脉粥样硬化碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)、转化生长因子β1(transforming growth factorβ1,TGFβ1)、胶原I、MMP1及TIMP1的表达。结果:2种药物均可显著降低兔血脂,辛伐他汀效果更强;2种药物均可显著消退动脉粥样硬化;药物治疗组bFGF、TGFβ1、胶原I、MMP1的mRNA表达水平均下降,其中辛伐他汀组的bFGF表达差异有统计学意义(P<0.05)。结论:2种药物均有消退动脉粥样硬化的作用, 药物通过减少生长因子和MMP1的表达,相对减少MMP1/TIMP1表达比例,起到稳定斑块的作用。

Abstract: To study the mRNA expression of basic fibroblast growth factor (bFGF), transforming growth factorβ(TGFβ1), collagen I matrix metalloproteinase 1 (MMP1) and its tissue inhibitor 1 (TIMP1) in atherosclerotics treated with QUYU XIAOBAN capsule and simvastatin. Methods: Atherosclerosis was induced in 40 rabbits by cholesterol feeding, and then QUYU XIAOBAN capsule and simvastatin were used for them. The tissues were detected by RTPCR, pathology and immunohistochemistry staining. Results: The lipids and the atherosclerotic plaque were decreased in both groups, but the former was more significantly in the simvastatin group; Whereas the mRNA level of basic fibroblast growth factor(bFGF), transforming growth factorβ(TGFβ1), collagen I, MMP1 and TIMP1 decreased more significantly in QUYU XIAOBAN capsule than in simvastatin group. Conclusion: Chinese medicine and simva statin can make the atherosclerotic plaque regression and stabilize the plaque by reducing the mRNA expression of bFGF,TGFβ1,collagen I and MMP1.
