辣椒素对膀胱癌细胞侵袭转移的影响

Effects of capsaicin on the invasion and metastasis of bladder cancer

摘要: 目的 探讨辣椒素对膀胱癌细胞系253J与RT112侵袭转移的影响。 方法 以0、100、 200 μmol/L辣椒素分别作用于人膀胱癌253J与RT112细胞0、12、24 h,采用MTT法检测253J与RT112细胞活力,划痕实验检测253J与RT112细胞迁移能力,Transwell法检测253J与RT112细胞侵袭能力。Western blotting法检测上皮间充质转化相关蛋白(E-钙黏蛋白,N-钙黏蛋白,波形蛋白)和Nrf2相关蛋白的表达水平。 结果 与0 μmol/L比较,在100、 200 μmol/L辣椒素作用下,253J与RT112各组细胞活性降低(P<0.01),侵袭能力下降(P<0.01),迁移能力也下降(253J: P<0.01; RT112: P=0.026或P=0.014)。细胞E-钙黏蛋白、C-nrf2表达上升,而波形蛋白与N-nrf2蛋白表达减弱, 并呈剂量-时间依赖性。 结论 辣椒素可抑制人膀胱癌细胞的侵袭与转移,该作用可能与Nrf2的调节有关。

Abstract: Objective To explore the inhibitory effect of capsaicin on the invasion and metastasis of human bladder cancer 253J and RT112 cells. Methods Human bladder cancer 253J and RT112 cells were treated with 0, 100, 200 μmol/L capsaicin for 0 h, 12 h and 24 h. The viability, migration and invasion of cells were measured with MTT assay, wound healing assay, and Transwell assay, respectively. The expressions of epithelial-mesenchymal transition(EMT)markers(E-cadherin, vimentin, and N-cadherin)and nuclear erythroid-related factor 2(Nrf2)proteins were analyzed with Western blotting. Results Compared with 0 μmol/L, in the 100 and 200 μmol/L capsaicin groups, the inhibition rate of cell viability increased(P<0.01), the index of invasion decreased(P<0.01), and migration decreased(253J: P<0.01; RT112: P=0.026 or P=0.014). The expressions of E-cadherin and C-nrf2 protein increased, while the expressions of vimentin and N-nrf2 protein decreased in a dose- and time-dependent manner. Conclusion Capsaicin can inhibit the invasion and metastasis of human bladder cancer cells possibly due to mediation of Nrf2 activity.
