葡萄籽原花青素对兔动脉粥样硬化氧化应激的影响

Oxidative effect of grape seed proanthocyanidin extract on atherosclerosis in rabbits

摘要:

目的    通过观察葡萄籽原花青素对动脉粥样硬化兔血浆丙二醛及氧化低密度脂蛋白及主动脉形态学的影响,探讨葡萄籽原花青素对氧化应激及动脉粥样硬化的预防作用。方法    将24只雄性新西兰白兔随机分为对照组、模型组及葡萄籽原花青素干预组,每组8只,分别给予不同的饲料,0、4、8、12周末留取血浆,使用酶联免疫吸附法检测血浆丙二醛及氧化低密度脂蛋白含量,留取主动脉行HE染色及扫描电镜观察。 结果    12周末氧化低密度脂蛋白含量模型组较对照组明显升高[(54.26±5.33)mg/L vs (24.48±3.13)mg/L, P﹤0.01],而葡萄籽原花青素组为(45.46±4.01)mg/L,与模型组差异具有统计学意义(P﹤0.01);模型组丙二醛的含量较对照组明显升高 [(7.49±0.53) μmol/L vs (3.08±0.13)μmol/L, P﹤0.01],而葡萄籽原花青素组含量为(6.90±0.59)μmol/L,与模型组差异具有统计学意义(P﹤0.05)。形态学观察可见,葡萄籽原花青素干预治疗后,局部破坏减轻,小栓子形成受抑。 结论    葡萄籽原花青素具有显著抗氧化效果,可抑制低密度脂蛋白氧化,发挥抗动脉粥样硬化形成作用。

Abstract:

Objective    To evaluate oxidative and anti-atherosclerotic effects of grape seed proanthocyanidin extract(GSPE) in rabbits by detecting malondialdehyde(MDA) and oxidized low-density-lipoprotein-cholesterol(oxLDL).  Methods    24 male New Zealand rabbits were randomized into three groups (control, model  and GSPE group), with 8 rabbits in each group. Blood samples were drawn from the middle arteries of rabbits′ ear at different time points, i.e. before the experiment, the fourth, the eighth, and the twelfth weekend of the experiment. Blood samples were analyzed for oxLDL and MDA with ELISA method. Aorta were excised and observed  morphologically through HE staining and electron microscope.  Results    At the 12th weekend, the serum oxLDL and MDA  increased significantly in model group than control group (54.26±5.33)mg/L vs (24.48±3.13)mg/L, (7.49±0.53)μmol/L vs (3.08±0.13)μmol/L; all P﹤0.01). In GSPE group, oxLDL and MDA were (45.46±4.01)mg/L and (6.90±0.59)μmol/L, respectively(P﹤0.01). Morphologically, the progress of atherosclerosis was obviously alleviated by GSPE while GSPE group was better than model group.  Conclusion    GSPE has  antioxidative effect in rabbits and it can alleviates the development of atherosclerosis significantly by inhibiting the formation of oxLDL.
