高碘对大鼠凝血功能的影响

Effect of excess iodine on coagulation function in rats

摘要:

目的  建立高碘喂养的大鼠动物模型,观察不同浓度的高碘对大鼠凝血功能影响的剂量与效应关系。方法  实验分4组:①正常饲料+正常水组(NI);②正常饲料+10倍碘组(10HI);③正常饲料+50倍碘组(50?HI);④正常饲料+100倍碘组(100HI);每周称体重,饲养6个月,用乙醚麻醉,从眼球静脉取血,CA7000全自动凝血分析仪测定凝血酶原时间(PT-time)、凝血酶原时间国际标准化比值(PT-INR)、活化部分凝血活酶时间(APTT)、凝血酶时间(TT)、纤维蛋白原时间(Fbgtime)、纤维蛋白原浓度(Fbg-c)、抗凝血酶Ⅲ活性(AT3:A)和D-二聚体(D-dimer)。结果  与NI相比,10?HI组的大鼠PT-time、PT-INR、Fbg-time、TT、APTT和AT3:A数值降低(PT、PT-INR、Fbgtime、AT3:A:P<0.05;TT、APTT P<0.01),Fbgc数值升高(P<0.01);而50HI和100HI组的大鼠PT-time、PT-INR、Fbg-time、TT、APTT数值升高(50HI组的Fbg-time、TT -P-<0.05;50HI、100HI组的其它指标P均<0.01),Fbg-c数值降低(50?HI组:P<0.05,100HI组P<0.01)。结论  适量高碘对大鼠凝血功能有促进作用,但是随着碘浓度升高,凝血功能降低。

Abstract:

Objective  To observe the dose-effect relation between excess iodine and coagulation function in rats. Methods  Wistar strain rats were divided into four groups: the normal diet and water group(NI), the normal diet and 10-fold iodine group(10HI), the normal diet and 50-fold iodine group(50HI), andthe normal diet and 100-fold iodine group(100HI). Body weights were determined weekly. After 6 months, blood was taken from the eyeball vein and then PT-Time, PT-INR, APTT, TT, Fbg-Time, Fbg-c, AT3:A, and D-dimer were determined by the  Sysmex CA7000 automated blood coagulation analyzer. Results  PT, PT-INR, Fbg-time, TT, APTT, and AT3: A were decreased while the number of Fbg-c was significantly increased in the 10HI group compared with the NI group(PT, PT-INR, Fbgtime,AT3: A:P<0.05: TT, APTT, Fbg-c P<0.01) PT-time, PT-INR, Fbg-time, TT, and APTT significantly increased while Fbg-c significantly decreased in the 50I and 100I groups comparing with the normal control group (except for Fbg-Time, TT and Fbg-c in the 50HI group P<0.05;the other blood coagulation indexes in the 50?HI and 100?HI groups, P<0.01)Conclusion  Appropriate excess iodine can promote blood coagulation of rats, however, with an increase of concentration of iodine, blood hypocoagulability is decreased.
