山东汉族人群IL12B基因多态性与慢性

Association analysis of the IL12B gene polymorphisms with chronic obstructive pulmonary disease in the Chinese Han population

摘要: 目的探讨白细胞介素12B(IL12B)基因1188A/C位点多态性与山东汉族人群慢性阻塞性肺疾病(COPD)易感性的关系。方法采用PCRRFLP法对324例COPD患者和389例健康人IL12B基因1188A/C位点多态性进行分型,对等位基因及基因型分布进行统计学分析。结果IL12B基因1188A/C多态位点AA、AC、CC基因型频率在病例组中分别为0.333?3、0.478?4和0.188?3,在对照组中分别为0.357?3、0.475?6和0.167?1,A和C等位基因型的频率在病例组中分别为0.572?5和0.427?5,在对照组分别为0.595?1和0.404?9,该位点基因型和等位基因型分布在病例组与对照组P值分别为0.689,0.389,差异均无统计学意义。结论IL12B基因1188A/C位点多态性与山东汉族人群COPD发生无明显相关性。

Abstract: To explore the relationship between single nuclear polymorphism (SNP) of 1188A/C in the interleukin 12B gene and susceptibility of chronic obstructive pulmonary disease (COPD) in the Chinese Han population from Shandong Province. MethodsSNP 1188A/C of the IL12B gene was detected by polymerase chain reaction (PCR) based on the restriction fragment length polymorphism (RFLP) technique in 324 COPD subjects and 389 healthy controls. ResultsThe frequencies of genotypes AA, AC, and CC at polymorphism sites 1188A/C were 0.333?3, 0.478?4, and 0.188?3 respectively in the COPD group, while they were 0.357?3, 0.475?6, and 0.167?1 in the control group. Also, the A type allele frequencies were 0.572?5 and 0.595?1 in patients and controls, and the C type allele frequency was 0.427?5 and 0.404?9 in COPD patients and healthy controls. There were no significant differences in genotype frequencies (P value=0.689) or allele frequencies between the two groups (P=0.389). ConclusionNo significant association was found between IL12B 1188A/C polymorphism and COPD susceptibility in the Chinese Han population from Shandong Province.
