超声经颅辅助尿激酶溶栓治疗急性脑梗死动物的实验研究

Transcranial ultrasound-enhanced thrombolysis by urokinase for acute cerebral infarction in rats

摘要: 目的:观察超声经颅辅助尿激酶溶栓治疗急性脑梗死动物的效果。方法:采用自身血栓栓塞法制备大鼠急性脑梗死模型,静脉给予尿激酶,同时经颅行超声治疗,NSS评分评价大鼠治疗前后的神经功能,TTC染色测量脑梗死灶的体积,光学显微镜与透射电子显微镜观察病理改变。结果:大剂量尿激酶组与超声加小剂量尿激酶组治疗后24h,NSS评分均明显低于治疗前(P<0.01)与梗死组(P<0.01),梗死灶体积均显著小于梗死组(P<0.01),但两者之间差异无统计学意义(P>0.05),光学显微镜与电子显微镜均见神经元缺血损伤明显减轻。结论:超声经颅辅助尿激酶溶栓可减小梗死灶体积,减轻神经元的缺血损伤,促进急性脑梗死大鼠神经功能的恢复,并可减少尿激酶的用量。

Abstract: Objective: To observe the effect of transcranial ultrasound-enhanced thrombolysis by urokinase on acute cerebral infarction in rats. Methods: The animal model of acute cerebral infarction was made by injecting auto-thrombsis into the left middle cerebral artery. Intravenous urokinase and transcranial ultrasound were given at the same time. The functioning of the nervous system was measured by NSS. The infarct volume was measured after TTC staining. Pathological changes were observed by a light microscope and transmission electron microscope. Results: The NSS score of the large dose urokinase group and the ultrasound plus small dose urokinase group at 24 hours after treatment was lower than that before treatment(P<0.01) and of that of the infarct group(P<0.01). The infarct volume of the two groups was lower than that of the infarct group, but there were no differences between the two groups(P>0.05). Remission of pathology was seen under both light microscope and transmission electron microscope. Conclusions: Transcranial ultrasound enhanced thrombolysis by urokinase can accelerate the recovery of the nervous system in acute cerebral infarction rats, diminish the infarct volume, decrease the ischemic changes of the neurons, and reduce the dosage of urokinase.
