Protective effect of oyster extract on apoptotic cells in neural tube defect induced by hyperthermia

摘要: 目的:探讨牡蛎提取物对高温致神经管畸形中凋亡细胞的保护作用。方法:取36只金黄地鼠孕鼠,随机分为4组:高温对照组和牡蛎保护Ⅰ、Ⅱ、Ⅲ组,用核固红-结晶紫染色方法,观察各组鼠胚的神经上皮和周围间充质细胞的凋亡状况。用免疫组织化学法和图像分析技术对各组鼠胚的神经上皮和周围间充质细胞凋亡相关蛋白Caspase-3的表达进行定性、定位和半定量观察。结果:①高温后8h、16h,对照组鼠胚神经上皮和周围间充质细胞凋亡数较多,牡蛎保护组细胞凋亡数明显减少(P<0.05, P<0.01);②高温后8h、16h,对照组鼠胚神经上皮和周围间充质内可见Caspase-3阳性表达细胞,牡蛎保护组的相应部位Caspase-3呈弱阳性或阴性表达(P<0.05)。结论:牡蛎提取物对高温致神经管畸形中神经上皮及周围间充质的凋亡细胞具有一定的保护作用。

Abstract: Objective: To explore the protective effect of oyster extract on apoptotic cells in neural tube defect(NTD) induced by hyperthermia. Methods: Thirty-six pregnant golden hamsters were divided into 4 groups at random: a control group subjected only to hyperthermia and oyster Ⅰ, Ⅱ and Ⅲ groups, with oyster extract. The nuclear fast redgentian violet staining method was used to observe the histological and the quantitative alteration of apoptosis cells during the development of the neural tube in each embryo. An immunohistochemistry method and image analysis were employed to explore the expression of Caspase-3 in the neuroepithelial and mesenchyme cells of each embryo. Results: ① At 8 and 16 hours after hyperthermia, compared with the control groups, the apoptosis index of neuroepithelial and mesenchymal cells in the oyster-treated groups decreased (P<0.05, P<0.01); ② At 8 and 16 hours after hyperthermia, the expression of Caspase-3 in the neuroepithelial cells was weaker in the oyster-treated than in the control groups (P<0.05). Conclusion: Oyster extract has a possible protective effect on the apoptosis cells of neuroepithelia and mesenchyme in NTD induced by hyperthermia.
