环氧化酶-抑制剂对白血病HL-60细胞的影响

Antiproliferation and apoptosis inducing of cyclooxygenase2(Cox-2)

摘要: 研究环氧化酶2(COX2)抑制剂塞来考昔(Celecoxib)对急性白血病HL60细胞的增殖抑制、诱导凋亡作用及其机制。方法:以不同浓度的Celecoxib作用于体外培养的HL60细胞,MMT法检测细胞生长抑制率,流式细胞仪检测细胞凋亡率,应用Hoechst 33258 荧光染色法观察细胞凋亡时的形态学变化,并对细胞凋亡前后的Caspase3活性进行检测。 结果:COX2抑制剂 Celecoxib可显著的抑制细胞的生长,并诱导细胞发生凋亡,在细胞凋亡过程中Caspase3活性显著升高,并呈现出明显的量-效与时-效关系。结论:COX2抑制剂 Celecoxib对HL60细胞具有显著的增殖抑制及诱导凋亡作用,升高Caspase3活性可能是其重要作用机制之一,这为COX2抑制剂进一步用于临床治疗白血病提供了有利的实验依据。

Abstract: To investigate the effect of cyclooxygenase2 (Cox2) inhibitor on leukemia HL60 (human acute promylocytic leukeamia cell line) cells. Methods: HL60 cells in culture medium were treated with different concentrations of Cox2 inhibitor, Celecoxib in vitro. MTT assay was used to detect the cell inhibition effect, and flow cytometry (FCM) was used to explore the cell apoptosis. The morphology of cell apoptosis was observed by Hoechst 33258 staining, and the activity of Caspase3 was detected before and after apoptosis. Results: Celecoxib revealed significant antiproliferation and apoptosis inducing effects on HL60 cells in vitro, and the activity of Caspase3 was upregulated remarkably in time and dosedependent manner. Conclusion: Celecoxib has significant antiproliferation and apoptosis inducing effects on HL60 cells, which may closely relate to the increasing of Caspase
