豚鼠透镜诱导性近视眼巩膜细胞凋亡的研究

摘要: 目的:探讨细胞凋亡在透镜诱导豚鼠近视眼后极部巩膜组织中的表达及其在近视形成中的作用。方法:选健康4周龄豚鼠45只,随机分成3组,1组(右眼配戴20D)、2组(右眼配戴10D)3组(不作任何干预)。实验前后用睫状肌麻痹下验光、A超动态观察豚鼠眼屈光度和眼轴长度,并取后极部巩膜组织,用电镜、终末脱氧核糖核苷酸转移酶介导原位缺口末端标记法(TUNEL)及流式细胞术检测巩膜的凋亡细胞。结果:实验后诱导眼形成近视,且伴有眼轴延长。并在诱导眼巩膜细胞中检测到凋亡细胞,凋亡细胞指数(AI)和凋亡率显著高于对照组(P<0.05),细胞增生指数(PI)显著低于对照组(P<0.05) 。结论:在透镜诱导豚鼠实验性近视眼后极部巩膜组织中不仅有细胞增殖的降低,而且还存在细胞凋亡的表达增多。

Abstract: To investigate the apoptosis cell in posterior sclera and to evaluate the apoptotic and the proliferative changes in lensinducted myopia. Method: A -20 D concave or -10 D concave was worn over the right eye in 30 fourweekold pigmented guinea pigs. The eyes were examined periodically with retinoscopy, and ascan ultrasonography. Two weeks later, the eyeballs were removed and the apoptosis cells were determined by electron microscopy, TUNEL technique and flow cytometry in sclera. Result: All eyes treated with -10.00 D or -20.00 D concave lens developed defocusinduced myopia. And axial lengths in lensinduced eyes remarkably increased in comparison with its control eyes. Apoptosis cells were observed in fibroblasts of sclera in the lensinduced eyes. Both the value of apoptotic index and the apoptosis rate in lensinduced eyes were significantly higher than the control group(P<0.01), and the value of proliferative index was lower than the control group(P<0.01). Conclusion: With the decreasing of the proliferation, the apoptosis cell increases in the posterior scleral fibroblasts in lensinduced myopia eye.
