干扰素和黄芪对心肌炎小鼠心肌细胞凋亡的影响

摘要: 方法:BALB/c小鼠腹腔接种半数组织感染量(TCID50)为108/ml的柯萨奇B3病毒0.1?ml。干扰素治疗组于接种病毒后24?h起腹腔注射α1b干扰素104IU/g体重,1次/d,连用5?d。黄芪治疗组于接种病毒后24?h起灌喂黄芪颗粒30?mg/g体重,1次/d,共5?d。接种病毒后第7、14、21天分别处死小鼠,原位TUNEL法检测心肌细胞凋亡,ELISA法测定血清肿瘤坏死因子α含量。结果:病毒性心肌炎小鼠心肌细胞凋亡百分率明显增高,血清肿瘤坏死因子α含量增加,干扰素和黄芪治疗可降低心肌细胞凋亡百分率及血清肿瘤坏死因子α含量。结论:α干扰素和黄芪有抑制心肌细胞凋亡作用。

Abstract: To investigate the influence of interferonα and astragalus membranaceus on cardiomyocyte apoptosis in murine viral myocarditis. Methods: 108TCID50/ml Coxsackievirus B3, 0.1?ml, was inoculated intraperitoneally in BALB/c mouse as model of murine viral myocarditis. Astragalus membranaceus granule was administrated by mouth in a dose of 30?mg/g/d in astragalus membranaceus granule treatment group, and interferonα1b was injected intraperitoneally in a dose of 104IU/g /d for 5 days since 24 hours after viral inoculation in interferonα treatment group. Mice were killed respetively on day 7,14, and 21 after inoculation. Cardiomyocyte apoptosis and serum TNFα concentration were determined by TUNEL in situ and ELISA respectively. Results: Percentage of cardiomyocyte apoptosis and concentration of serum TNFα increased in mice with viral myocarditis, and both of them reduced in interferonα and astragalus membranaceus granule treatment groups. Conclusion: Both interferonα and astragalus membranaceus can inhibit excessive cardiomyocyte apoptosis in murine viral myocarditis.
