CTLA-4Ig抑制再生障碍性贫血患者T淋巴细胞

摘要: 研究细胞毒性T淋巴细胞相关抗原4融合蛋白(CTLA4Ig)诱导再生障碍性贫血(AA)患者骨髓T淋巴细胞免疫耐受的作用。方法:采用免疫荧光标记法、流式细胞术分析CTLA4Ig阻断前后AA患者骨髓T淋巴细胞表型CD3+、CD4+、CD8+、CD25+、CD4+CD25+和CD8+CD25+的变化。结果:再障试验组与对照组相比,重型再障(SAA)和非重型再障(NSAA)患者骨髓CD3+、CD25+比值、CD4+和CD8+细胞中CD25+细胞比值均明显减低(P<0.05),CTLA4Ig对重型再障的抑制作用高于非重型再障,对CD4+细胞的抑制作用高于CD8+群体。结论:CTLA4Ig能够抑制再生障碍性贫血患者骨髓局部的异常自身免疫反应,降低骨髓T淋巴细胞对丝裂原的异常反应。

Abstract: o investigate the inhibition of the PHAinduced abnormal bone marrow T lymphocyte proliferation and activation by the CTLA4Ig fusion protein in vitro in patients with aplastic anemia (AA). Method: Flow cytometry and immunofluorescence were used to analyze the mononuclear cells from bone marrow of the aplastic anemia patients. Result: In contrast to the controls, the expressions of the CD3, CD25, CD25 on CD4+, CD8+ lymphocytes were statistically decreased(P<0.05) in both serious and nonserious AA. Conclusion: The CTLA4Ig fusion protein can induce the T cell tolerance and inhibit the T cell abnormal proliferation and activation in patients with AA.
