To investigate the expression of PTEN and MDM2 in the pathogenesis and prognosis of renal clear cell carcinoma. Methods: The expression of PTEN and MDM2 protein were evaluated in 58 renal clear cell carcinoma tissues, 15 renal tissues adjacent to carcinoma tissues and 10 normal renal tissues by immunohistochemistry method (SABC method). Results: PTEN was expressed in 34.4%(20/58)of in all the renal clear cell carcinoma patients, which was statistically significant lower than that in renal tissues adjacent to carcinoma tissues (86.6%, 13/15) and in the normal renal tissues (90%, 9/10) (both P<0.01), while it was not markedly different in renal tissues adjacent to carcinoma tissues and the normal renal tissues(P>0.05). PTEN expressed in StageⅠand Ⅱ renal carcinoma tissues (72.7%, 16/22) was much higher than that in Stage Ⅲ and Ⅳ carcinoma tissues (11.1%, 4/36). MDM2 was expressed in 68.9%(40/58)of all the renal clear cell carcinoma patients, which was statistically significant higher than that in renal tissues adjacent to carcinoma tissues (0.06%, 1/15) and the normal renal tissues (0, 0/10) (both P<0.01), while it was not significantly different in renal tissues adjacent to carcinoma tissues and the normal renal tissues(P>0.05), MDM2 expressed in Stage Ⅰ and Ⅱ renal carcinoma tissue (45%, 10/22) was markedly lower than that山东大学学报(医学版)44卷9期

in Stage Ⅲ and Ⅳ carcinoma tissue (83.3%, 30/36). The expression of PTEN was highly related with that of MDM2. Conclusions: The deletion or weak expression of PTEN and overexpression of MDM2 play a certain role in the occurrence of primary renal clear cell carcinoma and closely correlate with the development of renal carcinoma and lymphatic metastasis,which indicates that PTEN and MDM2 can be valued as biological indexes for development and prognosis of primary renal clear cell carcinoma.
