    Expression of PTEN and MDM2 protein in renal clear cell carcinoma

    To investigate the expression of PTEN and MDM2 in the pathogenesis and prognosis of renal clear cell carcinoma. Methods: The expression of PTEN and MDM2 protein were evaluated in 58 renal clear cell carcinoma tissues, 15 renal tissues adjacent to carcinoma tissues and 10 normal renal tissues by immunohistochemistry method (SABC method). Results: PTEN was expressed in 34.4%(20/58)of in all the renal clear cell carcinoma patients, which was statistically significant lower than that in renal tissues adjacent to carcinoma tissues (86.6%, 13/15) and in the normal renal tissues (90%, 9/10) (both P<0.01), while it was not markedly different in renal tissues adjacent to carcinoma tissues and the normal renal tissues(P>0.05). PTEN expressed in StageⅠand Ⅱ renal carcinoma tissues (72.7%, 16/22) was much higher than that in Stage Ⅲ and Ⅳ carcinoma tissues (11.1%, 4/36). MDM2 was expressed in 68.9%(40/58)of all the renal clear cell carcinoma patients, which was statistically significant higher than that in renal tissues adjacent to carcinoma tissues (0.06%, 1/15) and the normal renal tissues (0, 0/10) (both P<0.01), while it was not significantly different in renal tissues adjacent to carcinoma tissues and the normal renal tissues(P>0.05), MDM2 expressed in Stage Ⅰ and Ⅱ renal carcinoma tissue (45%, 10/22) was markedly lower than that山东大学学报(医学版)44卷9期 万芙荣,等.抑癌基因PTEN、癌基因MDM2在肾透明细胞癌中的表达及意义 in Stage Ⅲ and Ⅳ carcinoma tissue (83.3%, 30/36). The expression of PTEN was highly related with that of MDM2. Conclusions: The deletion or weak expression of PTEN and overexpression of MDM2 play a certain role in the occurrence of primary renal clear cell carcinoma and closely correlate with the development of renal carcinoma and lymphatic metastasis,which indicates that PTEN and MDM2 can be valued as biological indexes for development and prognosis of primary renal clear cell carcinoma.

    To construct a bispecific single chain antibody fragment antiCD3×antiTP3 diabody with dual specificity for both the human osteosarcoma cell antigen TP3 and CD3εchain of CD3/TCR. Methods: The expression plasmid pKIDCD3 × TP3 was constructed with gene of antibody variable fragment, and then it was transinfected into the E. coli K12 strain XL1Blue to obtain diabody. The diabody was purified by IMAC and its affinity was determined by flow cytometry and PAGE, at last, it was incubated with human T cell and osteosarcoma cell. Results: The diabody was absolute and stable in human plasma with high specificity for human T cells and osteosarcoma cells, and it also makes T cells approach target cells and lyse them. Conclusion: The diabody not only stimulates and retargets CTLs to osteosarcoma cells but also lyses them.

    To investigate the feasibility of threedimensional dynamic contrastenhanced MR angiography (3DDCEMRA) with realtime fluoroscopic triggering in carotid arteries. Methods: Threedimensional time of flight MR angiography (3DTOFMRA) and 3DDCEMRA were performed in 60 cases. 3DTOFMRA imaging sequence was threedimensional fast gradient echo with thin slice and multislab, and 3DDCEMRA imaging sequence was threedimensional time of flight spoiled gradient echo. The 2025?ml GdDTPA was injected with the rate of 2.03.0?ml/sec and the dose 0.150.2?ml/kg. With the MR fluoroscopic triggering technique, all the primary graphs were analyzed by maximum intensity projection reconstruction in the post processing station. The image qualities, included sensitivity, continuity and sharpedged degree of these two different techniques were evaluated according to the diagnosed demand and dissecting. Results: Major branch was displayed in 57 cases, which presented 95% of total of cases classified as good image quality. 3DDCEMRA had significantly higher sensitivity, continuity, and sharpedged degree than 3DTOFMRA in evaluation of carotiarteries. Conclusion: 3DDCEMRA is an effectively and minimally invasive method for evaluating lesions in carotid arteries.

    To observe the number and distribution of langerhans cells(LCs) and the expression of HLADR antigen in oral licen planus(OLP) and normal control buccal mucosa, and therefore to explore the pathogenesis of OLP. Methods: Polyclonal antibody to S100 protein and monoclonal antibody to HLADR antigens were applied to paraffinembedded sections by SABC immunohistochemical method in 25 cases of OLP and 8 cases of normal control. Results:LCs were identified with anitS100 and antiHLADR. In normal buccal mucosa, LCs mainly located in the basal cell layer, occasionally in the upper basal layer; whereas in OLP, they mainly located in the middle and lower regions of the prickle cell layer and the basal cell layer, but throughout the prickle cell layer in some. S100+ LC and HLADR+ LC significantly increased in the epithelia of OLP (P<0.001), and 56% of the OLP specimens(14/25)showed HLADR+ keratinocytes. There was correlation between the severity of the lymphatic infiltration in lamina propria and the basal cell degeneration in OLP, and more severe the lymophatic infiltration was, more severe the basal cell degeneration was. Conclusions: Presenting specified antigen to cytotoxic T cell, LCs induce the degeneration of keratinocytes. Thecellular immunity mediated by cytotoxic T cells may play a major role in the pathogenesis of OLP, and therefore LC may play an important role for OLP.

    To investigate the proliferation of endometrial stromal cells in endometriosis by interleukin1beta(IL1β). Methods: The eurotopic endometrial stromal cells of endometriosis cultured within 3~5 passages were treated with IL1β in a dosage of 0 to 10.0?ng/ml, and then the cell proliferation and the cell cycle were measured by MTT and flow cytometry (FCM) respectively. Results: The endometrial stromal cells had been growing luxuriantly, and G0/G1 phase cells were few than G2/M and S phase cells. Moreover, the proliferation was the most prominent when the cells were incubated in the presence of IL1β at 1.0?ng/ml concentration for 12 hours. Conclusions: IL1β induces the proliferation of endometrial stromal cells of endometriosis and promotes the transformation of G0/G1 phase cells into G2/M and S phase.

    To investigate the effect of cyclooxygenase2 (Cox2) inhibitor on leukemia HL60 (human acute promylocytic leukeamia cell line) cells. Methods: HL60 cells in culture medium were treated with different concentrations of Cox2 inhibitor, Celecoxib in vitro. MTT assay was used to detect the cell inhibition effect, and flow cytometry (FCM) was used to explore the cell apoptosis. The morphology of cell apoptosis was observed by Hoechst 33258 staining, and the activity of Caspase3 was detected before and after apoptosis. Results: Celecoxib revealed significant antiproliferation and apoptosis inducing effects on HL60 cells in vitro, and the activity of Caspase3 was upregulated remarkably in time and dosedependent manner. Conclusion: Celecoxib has significant antiproliferation and apoptosis inducing effects on HL60 cells, which may closely relate to the increasing of Caspase

    To prepare and evaluate the quality of Picamilon sodium dispersible tablets. Methods: Different disintegrants were screened and the best formulation was optimized by orthogon experiment design using the disintegration time as index, and the best formulation of Picamilon sodium dispersible tablets was authenticated by the initial stability experiment. Results: The best formulation of Picamilon sodium dispersible tablets was composed of Picamilon sodium 50mg, brushite 120?mg, Ldisplacehydroxypropyl cellulose and sodium carboxymethyl starch 16mg and 2% polyvidone k30 queous solution quantity sufficient. The effect of disintegration was very optimized, Ldisplacehydroxypropyl cellulose and sodium carboxymethyl starch were used in coordination with exterior addition and interior addition. The result showed that the disintegration time of optimized prescription formulation was 95s, and the dissolution percent at 10min of it was obviously super to that of the conventional tablets(P<0.05), and the quality of the dispersible tablets was very well by stability test. Conclusion: The dispersible tablets of Picamilon sodium have a higher dissolution speed than conventional tablet.

    To study three morphological temozolomide (TM) capsules′ dissolutions and absolute bioavailabilities in rabbits. Methods: The dissolution of differently morphological TM capsules was determined by UV spectrophotometry. The pharmacokinetics and bioavailability of three morphologies (A, B and C) in rabbits were studied. Results: The results from Weibull′s analysis showed that the dissolutions of TM capsules were very different, the dissolution rate of TM(C) was the fastest and that of TM(B) was the slowest; The bioavailability of TM(A) and TM(B) were bioequivalent (P>0.05), but that of TM(C) was obvious lower than the others (P<0.01). Conclusion: TM capsule A and B are bioequivalent and TM(C) is lower in the bioavailability in rabbits.

    To investigate the relation between the single nucleotide polymorphisms(SNPs) of tumor necrosis factorα(TNFα) or interleukin6(IL6) promoter genes and susceptibility to the patients with gastric adenocarcinoma or the patients with gastric adenocarcinoma with helicobacter pylori(HP) infection. Methods:The SNPs of the TNFα(238G/A and 308G/A) and IL6(597G/A,174G/C and 572G/C) were determined by gene chip in 65 patients with gastric adenocarcinoma and 71 healthy controls. The sera concentrations of IgG, IgM and IgA of HP antibodies were measured by ELISA. Results:H. pylori infection was detected in 69.2% of 65 patients and 46.5% of 71 controls (P=0.007, odds ratio [OR]=2.53,95% confidence interval [95%CI]=1.28-5.24). Frequencies of TNFα238GA genotype and A allele in patients with gastric adenocarcinoma were significantly higher than those in healthy controls(P=0.024, OR=2.44,95%CI=1.11-5.37 and P=0.039,OR=2.13,95%CI=1.03-4.41, respectively). Frequencies of TNFα238GA genotype and A allele in patients with gastric adenocarcinoma with HP infection were significantly higher than those in HPnegtive patients with gastric adenocarcinoma(P<0.05,OR=4.53, 95%CI=1.16-17.68 and P<0.05,OR=3.52, 95%CI=0.9812.64,respectively). The proportion of individuals carrying the IL6572CC genotype was significantly lower in patients with gastric adenocarcinoma than in the controls(P=0.014, OR=0.17, 95%CI=0.04-0.81). No association was found between any of the other polymorphisms and patients with gastric adenocarcinoma or patients with gastric adenocarcinoma with HP infection. Conclusion:There are significant correlation between TNFα238GA genotype and A allele and gastric adenocarcinoma or gastric adenocarcinoma with HP infection. IL6572CC genotype is a protective function against cancers.
    Effect of vWF, GMP140 of ultrafine particle of leeches in treating

    To estimate the value of ultrafine particle of Leeches on acute brain infarction disease. Methods: A total of 63 cases of early brain infarction disease were divided into 3 groups: group A,treated with normal medicine; group B, treated with normal medicine and bigger particle of Leeches;group C , treated with normal medicine and ultrafine particle of Leeches. The clinical symptom before and after treatment was observed, and also the level of blood Von Wille brand factor(vWF), pselection(GMP140) were observed. Results: The total response rate of group C and B was higher than that of group A(P<0.01, 0.05). The curative and marked response rates of group C were higher than those of group B(P<0.05). The improvement of blood vWF, GMP140 of group C was better than that of group B(P<0.05,0.01); and that of group B was better than group A(P<0.05, 0.01). Conclusion: There are serious vascular endothelium injury, platelet activity in both brain infarction patients. Ultrafine particle of Leeches is more effective in treating brain infarction disease than bigger particle of Leeches, and it is more effective in improving the blood levels of vWF, GMP140.
    Correlation between chemosensitivity and prognosis andabnormal expression of cyclin D1 and p27 in

    To prognose the chemosensitivity and the survival in terms of the expression levels of cyclin D1 and p27 protein in nonsmall cell lung cancer(NSCLC). Method: Enhanced immunohistochemical staining(SP) technique was used to detect the expression of cyclin D1 and p27 protein in 37 cases of primary NSCLC specimens paraffinembedded. Results:Of the 37 NSCLC cases, positive cyclin D1 was found in 21(56.76%), and there were significant correlation between response to chemotherapy, survival and the expression of cyclin D1(P<0.05). The positive expression rate of p27 protein was 32.43%(12/37), and there were no significant relationship between the expression of p27 and response to chemotherapy(P>0.05). The expression of p27 was significantly associated with the survival of NSCLC. Multivariate analysis showed that cyclin D1 was a good prognostic factor in NSCLC. Conclusion: Expression of cyclin D1 may be a good prognostic factor for chemosensitivity and survival in NSCLC. Furthermore, the expression of p27 may play a prognostic role in NSCLC.

As a supplement of conventional CT, CT perfusion imaging can reveal the perfusion informations of tissue or organs, such as hemodynamics, microcirculation status. It is helpful not only in finding morphologic changes, but also in providing functional and metabolizable information about the circulation parameters of hepatic diseases. So, it is valid for studying liver disorders by providing functional information, especial for the differential diagnosis and assessment for therapeutic response.
