Immunotherapeutic effect of dendritic cell vaccine modified with the CXCL10
gene and tumor cell RNA on mice with prostate cancer

Abstract: To evaluate the effect of therapeutic vaccine against prostate cancer based on dendritic cell (DC) vaccine modified with tumor cell RNA and the CXCL10 gene both in vitro and in vivo. MethodsMouse bone marrow DC was pulsed with cell RNA from the RM1 prostate cancer cell line and then transfected with the recombinant plasmid CXCL10／pcDNA31 by DOTAP liposome. The immunotherapeutic effects of DC vaccine on mice with prostate cancer were assessed. ResultsThe DC vaccine increased the preferential chemotaxis to T cells, and the mice immunized with DC vaccine exhibited a specific CTL response compared with other groups. In the C57BL／6 mice model with preestablished subcutaneous RM1 prostate cancer, immunization of DC vaccine inhibited tumor growth more significantly when compared with other counterparts and the survival time of micetreated with the DC vaccine was greatly extended. The DC vaccine immunized mice also exhibited an effective resistance to tumor challenge. ConclusionDC vaccine modified with CXCL10 gene and tumor cell RNA might elicit significant antitumor effects through efficient induction of antitumor immunity and might be of therapeutic potentials for prostate cancer.
