Abstract: To construct recombinant eukaryotic expression vector, pcDNA3.1NKX3.1 and express it in prostate cancer PC3 and LNCaP cells. Methods： The NKX3.1 complete decoding sequence was amplified by RTPCR by using RNA from LNCaP cell and was cloned into pCR2.1 vector by T/A ligation, and then into pcDNA3.1(+) vector to generate pcDNA3.1NKX3.1 which was instantaneously transfected into prostate cancer PC3 and LNCaP cells. The expression of NKX3.1 mRNA and protein was verified by RTPCR and Western blot. Results： The sequence of cloned NKX3.1 cDNA was identical to that documented in GeneBank. RTPCR and Western blot results indicated that NKX3.1 cDNA was effectually expressed in the transfected PC3 and LNCaP cells. Conclusion： The eukaryotic expression vector pcDNA3.1NKX3.1 is successfully constructed and effectually expressed in PC3 and LNCaP cells.
