Abstract: To investigate the effect of hypoxia on the expressions of MMPs and TIMPs in human breast carcinoma cell line MDA435. Methods： After the MDA435 cells being cultured under normoxia（21% O2, 5% CO2）or hypoxia（1% O2, 5% CO2 and 94% N2）for 6 hours, the expressions of MMP2, 9 and TIMP2, 1 were detected in mRNA level by RTPCR. Results: The expressions of MMP2 and TIMP2 were not observed in human breast cancer cell line MDA435； the expression of MMP9 increased significantly in mRNA levels when cells were cultured under normoxia (P＜0.05), but no significant differences of TIMP1 were observed under normoxia or hypoxia (P＞0.05). Conclusion: Hypoxia could affect the expression of MMP9 in human breast cancer cell line MDA435，indicating that in vitro study on the molecule mechanism of neoplasm metastasis should take the oxygen environment into conclusion.
