Abstract: To analyze the association of the estrogen receptor α (ERα) gene polymorphisms with the Th1/Th2 cytokine drift in systemic lupus erythematosus (SLE). MethodsGenomic DNA was extracted and polymorphisms of Xba Ⅰ and Pvu Ⅱ in intron 1 of the ERα gene were determined by the polymerase chain reactionrestriction fragment length polymorphisms (PCR-RFLP) method. The mRNA levels of IL-10, IL-4, IFN-γ and IL-2 were determined by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR). ResultsThe IL-10 and IL-4 mRNA expressions of SLE patients with the PpXx genotype were higher than those of controls (P＜0.01, P＜0.05), however the IFN-γ and IL-2 mRNA expressions of SLE patients with the Ppxx genotype were lower than those of controls (P＜0.05). The mRNA expression of four cytokines had no significant differences in various genotypes among the controls. ConclusionThe various ERα genotypes are relevant to cytokine expression in different individuals. The ERα gene polymorphisms may influence cytokine expression to some extent based on the gene background.
